OX40/OX40L分子在B6.MRL-Faslpr/J狼疮小鼠脾脏组织中的表达及意义

【摘要】 为了探讨OX40和OX40L分子在B6.MRL-Faslpr/J(简称B6.Fas)狼疮小鼠淋巴细胞上的表达及其在系统性红斑狼疮(systemic lupus erythematosus,SLE)发病机制中可能的作用,本课题采用流式细胞仪检测技术对小鼠脾脏T细胞上OX40和B细胞上OX40L的表达水平进行分析并采用Albustix试纸法检测小鼠24h尿蛋白量。结果显示,20周龄B6.Fas狼疮小鼠伴有异常升高的蛋白尿(++~+++)、28周龄B6.Fas狼疮小鼠蛋白尿含量进一步升高为++++,12周龄B6.Fas狼疮小鼠及正常小鼠均为≦+。与正常对照小鼠相比,12周龄B6.Fas狼疮小鼠T、B细胞上OX40/OX40L的表达无显著差异(P>0.05);20周、28周龄B6.Fas狼疮小鼠CD4+T细胞上OX40表达水平以及B细胞上OX40L表达水平明显升高(P<0.05),而CD8+T细胞上OX40表达水平无显著差异(P>0.05);并且28周龄B6.Fas狼疮小鼠T、B细胞上OX40/OX40L的表达水平高于20周龄小鼠(P<0.05)。这表明伴随蛋白尿量的增加,狼疮小鼠脾脏T细胞和B细胞上OX40和OX40L分子表达异常升高,提示OX40/OX40L信号可能与狼疮的发病以及病情发展有一定的相关性。

【Abstract】 We used B6.MRL-Faslpr/J(B6.Fas) mouse model of systemic lupus erythematosus(SLE) to investigate the expression of OX40 and OX40L molecules in spleen T lymphocytes and B lymphocytes and to explore their possible roles in the pathogenesis of SLE,OX40 and OX40L expression on T cells and B cells were detected by flow cytometry.Proteinuria was evaluated by Albustix dipstick.The results showed that,compared with the control group,while both the levels of OX40 and OX40L on T cells and B cells in 12 weeks old lupus mice had no significant change(P>0.05),but those on CD19+ B cells in 20 weeks old and 28 weeks old lupus mice were significantly upregulated(P<0.05),OX40 levels on CD8+ T cells had no significant change(P>0.05).The expression of OX40 and OX40L in the 28 weeks old lupus mice were higher than those in the 20 weeks old ones(P<0.05).In the lupus group,protein concentration in the uria was abnormally elevated with age and no proteinuria was found in the normal controls.OX40 and OX40L were abnormally expressed on CD4+ T cells and CD19+ B cells from the spleen of lupus mice with proteinuria,and the expression levels were increased in correlation with increased proteinuria,suggesting that OX40/OX40L signal may play a critical role in the development of SLE.
