IgA肾病大鼠骨髓内皮祖细胞的变化及意义

【摘要】 目的探讨IgA肾病大鼠骨髓内皮祖细胞(EPCs)在IgA肾病发生、发展中的意义。方法 SD大鼠20只随机分为IgA肾病组(IgAN组)和对照组各10只。参照文献采用牛血清白蛋白+脂多糖+四氯化碳的改良方法制备大鼠IgA肾病模型并鉴定。密度梯度离心法分离骨髓单个核细胞,体外培养为EPCs,流式细胞仪鉴定细胞表型(CD34、CD133、FLK-1)。激光共聚焦显微镜鉴定FITC标记的荆豆凝血素1(FITC-UEA-1)和Dil标记的乙酰低密度脂蛋白(DiI-acLDL),免疫荧光双阳性细胞为正在分化的EPCs。Transwell小室和CCK-8检测EPCs的迁移能力和增殖能力。结果 IgAN组骨髓EPCs数量较对照组明显减少,迁移和增殖能力均明显下降(P均<0.05)。Pearson相关分析显示,IgAN大鼠骨髓EPCs数量与血清BUN、Scr、IgA、24 h尿蛋白定量呈负相关(r分别为-0.49、-0.65、-0.81、-0.95,P均<0.05)。结论 IgA肾病的发生、发展可能与骨髓EPCs数量减少、迁移、增殖功能减退有关。

【Abstract】 Objective To investigate the potential role of bone marrow-derived endothelial progenitor cells( EPCs) in rats with primary immunoglobulin A nephropathy( IgAN). Methods Twenty SD rats were randomly divided into the IgAN group( n = 10) and the normal control group( n = 10). Mononuclear cells were isolated by Ficoll density gradient centrifugation from bone marrow. EPCs were seeded on the M199 culture plates. The cell phenotypes( CD34,CD133 and FLK-1) were detected by flow cytometry,and DiI-acLDL and FITC-UEA-1 were detected by Laser Confocal microscopy. The proliferation and migration of EPCs were measured by CCK8 assay and Transwell chamber assay,respectively. Results Compared with the normal control group,the number of EPCs was significantly reduced,and the abilities of proliferation and migration of EPCs were impaired in the IgAN group( P < 0. 05). Pearson correlation analysis showed that there were negative correlations between the number of EPCs and BUN,Scr,serum level of IgA and 24-hour urine protein( r =- 0. 49,- 0. 65,- 0. 81and- 0. 95,respectively; all P < 0. 05). Conclusion The reduction of the number and the dysfunction of migration and proliferation of EPCs may be correlated with the pathogenesis and development of IgAN.
