Akt在脑胶质瘤中的表达及其对脑胶质瘤细胞放射敏感性的影响

【摘要】 目的观察Akt基因在脑胶质瘤组织中的表达情况,并探讨其对脑胶质瘤细胞放射敏感性的影响。方法采用RT-PCR法、免疫组化法检测丝氨酸/苏氨酸蛋白激酶(Akt)基因在40份脑胶质瘤组织、6份脑正常组织中的表达情况,用LY294002阻断磷酯酰肌醇3激酶(PI3K)/Akt信号通路,MTT法检测脑胶质瘤细胞受X线照射后的增殖情况。结果在40份脑胶质瘤组织中,Akt基因的表达高于正常脑组织(P<0.05)。LY294002阻断PI3K/Akt信号通路后,可以明显地提高脑胶质瘤细胞对放射的敏感性(P<0.05)。结论 Akt基因的过表达与脑胶质瘤的发生和发展可能密切相关,阻断PI3K/Akt信号通路,可以提高脑胶质瘤细胞对放射的敏感性。

【Abstract】 Objective To observe the expression of Akt in glioma tissues and to investigate the effect of blocking PI3K / Akt signaling pathway on the proliferation of U87 cells induced by radiation. Methods The expression of Akt was detected by RT-PCR and immunohistochemistry in 40 cases of glioma tissues and 6 cases of normal brain tissues. LY294002 was used to block PI3K / Akt signaling pathway. The proliferation of glioma cells after radiation was determined by MTT assay. Results The expression of Akt in glioma tissues was higher than that in normal brain tissues( P < 0. 05). Inhibition of PI3K / Akt signaling pathway could significantly sensitize glioma cells to radiation( P < 0. 05). Conclusion The overexpression of Akt gene may be closely related to the occurrence and development of glioma,and blocking the PI3K / Akt signaling pathway can enhance the sensitivity of glioma cells to radiation.
