ALA-PDT对人食管癌Eca-109细胞相关凋亡蛋白表达的影响
Effect of ALA-PDT on related apoptosis protein expression in human esophageal cancer cell line Eca-109
【摘要】 目的探讨5-氨基乙酰丙酸—光动力疗法(ALA-PDT)对人食管癌Eca-109细胞相关凋亡蛋白表达的影响。方法将人食管癌Eca-109细胞经过培养、处理后分为两组,ALA-PDT组(PDT组)细胞用ALA孵育6 h后在光动力治疗系统下进行光照,光照后24 h提取细胞总蛋白和胞质蛋白;对照组细胞不进行光动力治疗。采用West-ern blot法检测两组细胞中的Bcl-2、Cytc蛋白表达。结果 ALA-PDT作用于人食管癌Eca-109细胞后,与对照组比较,PDT组胞质中的Bcl-2蛋白表达量明显降低,Cytc蛋白表达量明显升高(P均<0.05)。结论线粒体凋亡通路可能是ALA-PDT诱导Eca-109细胞凋亡的主要通路,Bcl-2可能为该凋亡通路的靶基因之一,释放Cytc进而诱导细胞凋亡可能为ALA-PDT诱导人食管癌Eca-109细胞凋亡的线粒体通路终末效应之一。
【Abstract】 Objective To investigate the effect of 5-aminolevulinic acid-mediated photodynamic therapy(ALA-PDT) on related apoptosis protein expression in esophageal cell line Eca-109.Methods The Eca-109 cells were divided into two groups after culture and treatment:PDT group and control group.The cells of PDT group were incubated with ALA for 6 h and irradiated under the 630 nm laser system,then the total cellular protein and cytoplasmic protein were extracted after 24h;whereas the control group without any treatment.Western Blotting assay was used to detect the protein expression changes of Bcl-2 and Cytc.Results After ALA-PDT,compared with the control group,the expression of Bcl-2 protein was significant down-regulation in the Eca-109 cells,while the expression of Cytc was significant up-regulation(all P<0.05).Conclusions Mitochondria-dependent apoptosis pathway may be the main apoptosis way,and Bcl-2 may be one of the target genes in ALA-PDT on Eca-109,which releases Cytc from mitochondria to induce apoptosis,and it may be one of the final reactions in the mitochondria-dependent apoptosis pathway.
