喷雾干燥对人溶菌酶转基因山羊乳汁的影响

【摘要】 【目的】揭示奶粉制备过程对转基因山羊乳汁中表达的重组人溶菌酶(recombinant human lysozyme,rhLYZ)的影响。【方法】对表达有2mg/mL rhLYZ的转基因山羊乳汁进行了喷雾干燥,对获得的奶粉样品进行了溶解性,乳汁成分,蛋白组分以及溶菌酶活性的测定,对测定结果进行统计分析。【结果】喷雾干燥引发非脂固形物的损失率为42.05%;整个热处理过程引发可溶性蛋白损失率为25.3%;乳汁中rhLYZ的损失为51.1%,扣除总干物质的损耗,喷雾干燥过程引发了约17.7%的rhLYZ生物学活性损失。【结论】制备奶粉过程对转基因山羊乳汁中表达的人溶菌酶影响较小,喷雾干燥是人溶菌酶羊奶的一种可行的保存和加工方式。

【Abstract】 【Objective】 The influence of milk powder preparation process on the physicochemical properties of transgenic goat milk and the recombinant human lysozyme(rhLYZ) were studied.【Method】 The transgenic goat milk with 2 mg/mL of rhLYZ was treated by spray-drying.The solubility,milk composition,protein fractions,and lysozyme activity of milk samples were tested.The results were statistical analyzed.【Results】 The lose rate of non-fat solids caused by spray-drying was up to 42.05%;the lose rate of dissolvable proteins was about 25.3%;and about 17.7% of rhLYZ biological activity was losted which resulted from the drying process.【Conclusion】 The influence of milk powder preparation process on rhLYZ was mild,and the milk powder could be a feasible method for preserving and processing rhLYZ milk.
