甲型H1N1病毒感染BEAS-2B细胞差异蛋白的初步研究
Study on the cellular differential proteins in BEAS-2B cells infected with 2009 (H1N1) influenza viruses
【摘要】 目的通过建立2009H1N1流感病毒感染BEAS-2B(人支气管上皮细胞)细胞模型,探讨病毒感染细胞后细胞蛋白质差异变化,为2009H1N1流感病毒的致病机理研究奠定基础。方法 100 TCID50 2009H1N1(D、S、O)流感病毒感染BEAS-2B细胞12、24、48、72h后提取细胞总蛋白进行双向电泳,Image MasterTM 2D Platinum Software 7.0软件分析图像,对有差异的蛋白酶解,质谱分析鉴定差异蛋白。结果质谱共鉴定出包括蛋白酶体α5、巨噬细胞移动抑制因子(MIF)、前纤维蛋白1(profilin-1)、α-2干扰素等13个差异蛋白。结论 13个差异蛋白可能参与了2009H1N1流感病毒的致细胞病变过程。
【Abstract】 Objective To elucidate the pathogenesis mechanism of 2009H1N1 influenza virus infection,2009H1N1 influenza virus infected-BEAS-2B cellular models were established,and analysis was performed to investigate alterations of protein expression in cells infected with virus.Methods BEAS-2B cells infected with 2009H1N1 influenza virus were established using 100 TCID 50 2009H1N1(D,S,O) influenza virus,the total cellular proteins were extracted over 12,24,48 and 72 h post-infection,total proteins were quantitated,and then 2D electrophoresis,images were analyzed by using Image Master TM 2D Platinum Software 7.0.The different proteins were enzymaticed and then indentified by MS.Results 13 different proteins were identified by MS.They included proteasome subunit alpha type-5,MIF,profilin-1,interferon alpha-2 and others...Conclusion These proteins may be involved in the process of the 2009H1N1 influenza virus cytopathic.
