【摘要】 目的探讨CXCR4 shRNA对胶质瘤细胞U251体外增殖和侵袭能力的影响,为CXCR4 shRNA治疗胶质瘤提供实验依据。方法用脂质体Lipofectimine TM2000将CXCR4-shRNA转染U251细胞,MTT实验检测转染细胞的增殖,Transwell侵袭实验检测转染细胞的迁移和侵袭能力。结果与转染非特异的pGPU6/GFP/NC组和未转染组比较,转染CXCR4-shRNA载体pGPU6/GFP/Rac1-421组的U251细胞CXCR4 mRNA显著降低(P<0.05),并且细胞体外增殖能力减弱(P<0.05),侵袭到滤膜底面的细胞数为36.67±9.76,分别低于对应的pGPU6/GFP/NC组和未转染组(P<0.05)。结论 CXCR4 shRNA载体pGPU6/GFP/Rac1-421可下调U251细胞CXCR4的表达,抑制U251细胞的增殖和迁移侵袭能力,提示CXCR4在胶质瘤发展过程中具有重要作用。更多还原

【Abstract】 Objective To investigate the effect of CXCR4 shRNA on glioma cell U251 proliferation and invasion in vitro and to provide experimental basis for treatment of glioma with CXCR4 shRNA.Methods CXCR4-shRNA was transfected into U251 cells using liposome LipofectimineTM2000,proliferation of transfected cells were detected by MTT experiment.The migration ability was detected by Transwell cells invasion experiment.Results CXCR4 mRNA of cells transfected with pGPU6/GFP/Rac1-421 in vector group decreased significantly than those U251 cells transfected with non pGPU6/GFP/NC group and non-transfection group(P<0.05),and their cell proliferation in vitro also decreased(P<0.05),the number of cells invaded the membrane was(36.67±9.76),respectively,lower than that in pGPU6/GFP/NC group and nontransfection group(P<0.05).Conclusions CXCR4 shRNA vector pGPU6/GFP/Rac1-421 can down-regulate CXCR4 mRNA expression of U251 cells,inhibit proliferation and migration U251 cells suggesting that CXCR4 plays an important role in the development of gliomas.更多还原
