频繁开花高产小桐子组织培养的初步研究
Tissue Culture of Frequently Flowering High Yield Jatropha curcas L.
【摘要】 为了奠定频繁开花型高产小桐子的快繁及转基因基础,通过对小桐子茎段腋芽培养、叶片和叶柄愈伤诱导及不定芽分化的不同激素组合培养进行研究。结果表明:适合该高产品种的茎段培养的最佳培养基为MS+6-BA0.5 mg/L+IBA 0.1 mg/L,叶片愈伤诱导及不定芽分化培养基为MS+6-BA 2.5 mg/L+IBA 0.5 mg/L,叶柄愈伤诱导及不定芽分化培养基为MS+6-BA 1.0 mg/L+IBA 0.2 mg/L,生根培养以1/2MS+IBA 0.5 mg/L+AC 0.1%较为适宜。引起小桐子组培苗玻璃化现象的主要原因之一是BA浓度过高;在不定芽的分化培养中,愈伤过多会严重影响不定芽的数量及质量。
【Abstract】 In order to provide a foundation for rapid propagation in vitro culture and transgenosis of frequently flowering of Jatropha curcas L., different combinations of phytohormone were used in the research of stem axillary bud culture, callus induction and differentiation of adventitious buds of leaf and petiole. Results showed that the optimal medium of the variety for stem culture was MS+6-BA 0.5 mg/L+ IBA 0.1 mg/L, for the callus induction and differentiation of adventitious buds of leaf was MS+6-BA 2.5 mg/L+IBA 0.5 mg/L, for the callus induction and differentiation of adventitious buds of petiole was MS+6-BA 1.0 mg/L+IBA 0.2 mg/L, and the 1/2 MS medium with 0.5 mg/L IBA and AC 0.1% was good for formation and growth of adventitious roots. One of the major causes for nitrification of Jatropha curcas was excessive concentration of BA; in addition, too many callus could seriously affect the quantity and quality of adventitious buds in the differentiation process.
