乳腺癌突变型p53与P-gp和GST-π及Topo-Ⅱ表达的相关性研究

【摘要】 目的:探讨突变型p53蛋白与P-gp、GST-π和Topo-Ⅱ3种耐药蛋白在乳腺癌组织中的表达特点及其临床意义。方法:免疫组化法检测260例乳腺癌中突变型p53与P-gp、Topo-Ⅱ和GST-π的表达情况。结果:乳腺癌中不同组织学类型与P-gp(χ2=3.84,P=0.049)和Topo-Ⅱ(χ2=7.75,P=0.005)表达差异有统计学意义;淋巴结的转移情况与P-gp(χ2=5.09,P=0.024)表达相关;临床分期与突变型p53(χ2=8.48,P=0.036)、P-gp(χ2=6.53,P=0.018)和Topo-Ⅱ(χ2=4.28,P=0.039)表达相关;与预后相关的因素包括突变型p53(χ2=4.26,P=0.039)、P-gp(χ2=19.25,P<0.001)和GST-π(χ2=5.55,P=0.019);突变型p53与P-gp(χ2=5.33,P=0.021)、突变型p53与GST-π(χ2=9.56,P=0.002)和突变型p53与Topo-Ⅱ(χ2=5.23,P=0.022)阳性表达之间均呈显著相关性。结论:突变型p53的表达可能在乳腺癌多药耐药的发生机制中起到了一定的作用。

【Abstract】 OBJECTIVE:To explain whether expression of mutant p53gene is correlated to the expressions of P-gp,GST-πand Topo-Ⅱin breast cancer.METHODS:SP immunohistochemical method was used to examine expressions of mutant p53,P-gp,GST-πand Topo-Ⅱin 260 samples of primary breast cancer.RESULTS:There were significantly difference between different histological types with P-gp(χ2=3.84,P=0.049)and Topo-Ⅱ(χ2=7.75,P=0.005);lymph node metastasis was correlated to P-gp(χ2=5.09,P=0.024);clinical stage was correlated to mutant p53(χ2=8.48,P=0.036),P-gp(χ2=6.53,P=0.018)and Topo-Ⅱ(χ2=4.28,P=0.039);and prognostic factors,including mutant p53(χ2=4.26,P=0.039),P-gp(χ2=19.25,P<0.001)and GST-π(χ2=5.55,P=0.019);There were significant correlations between mutant p53 and expressions of P-gp,GST-πand Topo-Ⅱin breast cancer(χ2=5.33,P=0.021;χ2=9.56,P=0.002;χ2=5.23,P=0.022).CONCLUSION:Expression of mutant p53may play certain role in the mechanism of multidrug resistance in breast cancer.
