TCDD和B[a]P诱导大鼠肺癌变过程CYPIA1表达变化的研究
Expression of CYPIA1 in the process of rat lung carcinogenesis induced by TCDD and B[a]P
【摘要】 目的:利用四氧二苯二氧杂环己二烯(TCDD,二噁英)和苯并(a)芘(B[a]P)致大鼠肺癌的动物模型,研究肺癌变过程中CYPIA1基因mRNA和蛋白水平的表达。方法:应用RT-PCR方法及荧光免疫法检测大鼠正常肺组织、癌前病变组织和肺癌组织中CYPIA1基因mRNA的表达及CYPIA1基因在肺组织蛋白水平的表达,评价CYPIA1基因在肺癌变过程的作用。结果:CYPIA1 mRNA/β-actin比值在TCDD、TCDD+B[a]P、B[a]P致癌组及对照组分别为0.956 4±0.328 2、1.153 3±0.432 6、0.252 4±0.103 2和0.093 6±0.032 7;在大鼠正常肺组织、正常增生、癌前病变和肺癌组织中分别为0.105±0.098、0.235±0.108、1.127±0.295和1.564±0.421。结果表明,CYPIA1mRNA在TCDD+B[a]P、TCDD致癌组有较强表达,在肺癌前病变和肺癌组织中有较强表达;CYP1A1蛋白表达灰度值在TCDD、TC-DD+B[a]P、B[a]P致癌组及对照组分别为41.76±4.82、46.28±4.32、31.02±3.16和18.75±2.27,在大鼠肺正常组织、正常增生、癌前病变和肺癌组织中分别为19.77±2.74、26.71±3.32、43.95±3.98和47.38±4.96。结果表明,CYP1A1蛋白在TCDD+B[a]P和TCDD致癌组有较强表达,在肺癌前病变和肺癌组织中有较强表达。结论:TCDD和B[a]P对CYPIA1有较好的诱导作用,CYPIA1高表达与肺癌的发生有着密切的关系,其在肺癌前病变和肺癌组织中的高表达,可能是肺癌前病变、肺癌发生的一个标志。
【Abstract】 OBJECTIVE:To investigate the role of gene CYPIA1 in the process of lung carcinogenesis.METHODS: In the animal model of rats lung cancer induced by dioxin and B[a]P,the level of CYPIA1 mRNA and CYPIA1 protein expression in groups of TCDD,TCDD+ B[a]P,B[a]P and control group were tested.RESULTS: The ratio of CYPIA1mRNA/β-actin in cancerogen groups of TCDD,TCDD+B[a]P,B[a]P and control group were 0.956 4±0.328 2,1.153 3±0.432 6,0.252 4±0.103 2,0.093 6±0.032 7;in normal epithelium,epithelial hyperplasia,atypical hyperplasia,primary carcinoma were 0.105±0.098,0.235±0.108,1.127±0.295,1.564±0.421 respectively.There was a high expression of CYPIA1 mRNA in group of TCDD,TCDD+ B[a]P,and high expression of CYPIA1 mRNA in atypical hyperplasia and primary carcinoma.The CYPIA1 protein Grey level in cancerogen groups of TCDD,TCDD+B[a]P,B[a]P and control group were 41.76±4.82,46.28±4.32,31.02±3.16,18.75±2.27,in normal epithelium,epithelial hyperplasia,atypical hyperplasia and primary carcinoma were 19.77±2.74,26.71±3.32,43.95±3.98,47.38±4.96.There was a high expression of CYPIA1 protein in groups of TCDD+ B[a]P and TCDD,high expression of CYPIA1 protein in atypical hyperplasia and primary carcinoma.CONCLUSIONS:Our resμlts suggest that TCDD and B[a]P can increase the expression of CYP1A1 gene.The lung cancer is correlation to the high expression of CYPIA1 and the high expression of CYPIA1 is an important biomarker for atypical hyperplasia and lunge cancer.
