慢性应激抑郁模型大鼠海马组织细胞色素C的表达及抗抑郁药物的干预作用
Expression of Cytochrome C in Hippocampus of Chronic Stress Depression Rats and the Interference of Antidepressants
【摘要】 目的观察慢性应激抑郁模型大鼠海马组织细胞色素C的表达及抗抑郁药物的干预作用。方法将雄性Wistar大鼠60只采用随机排列表法分为对照组、模型组、氟西汀组和噻萘普汀组,每组15只。模型制备期间对大鼠进行刺激前,氟西汀组大鼠采用氟西汀10.0 mg/kg灌胃,噻萘普汀组大鼠采用噻萘普汀50.0 mg/kg灌胃,对照组和模型组大鼠采用等体积的0.9%氯化钠溶液灌胃。采用蛋白印迹法测定各组大鼠海马组织细胞色素C的表达水平,以整合光密度值(IDV)表示;采用开场法测定各组大鼠模型制备前后5 min内的行为学观察指标,包括水平穿越格数、竖立次数、修饰次数,之后进行液体消耗实验,计算24 h内蔗糖水消耗百分比。结果对照组大鼠平均IDV值为(24 631±2 194),模型组为(38 963±5 384),氟西汀组为(27 220±2 892),噻萘普汀组为(33 204±4 174),4组大鼠细胞色素C表达水平比较,差异有统计学意义(F=33.343,P=0.000)。4组大鼠模型制备前水平穿越格数、竖立次数、修饰次数及蔗糖水消耗百分比比较,差异均无统计学意义(P>0.05);制备后比较,差异均有统计学意义(P<0.05)。结论慢性应激抑郁模型大鼠海马组织细胞色素C表达水平降低,抗抑郁药物可能通过抑制神经元内线粒体细胞色素C的释放以减少应激导致的神经元凋亡。
【Abstract】 Objective To investigate the expression of Cytochrome C in hippocampus of chronic stress depression rats and the interference of antidepressants.Methods 60 Wistar rats were randomly divided into control group,model group,fluoxetine group and tianeptine group with each group 15 rats.Before stimulation to the rats during model preparation,the fluoxetine group was given intragastric administration of 10.0 mg/kg fluoxetine,the tianeptine group was given intragastric administration of 50.0 mg/kg tianeptine,and the control group and model group were both given intragastric administration of 0.9% sodium chloride solution with the same volume.The expression of Cytochrome C was detected by Western Blot method and was expressed by IDV.Ethology indexes(including horizontal squares crossed,erection times and modification times)five minutes before and after model establishment were detected by open-field method,and experiment of fluid consumption was conducted to calculate the percentage of sacchar-consumption within 24 h.Results The average IDV was(24 631±2 194),(38 963±5 384),(27 220±2 892)and(33 204±4 174) respectively in control group,model group,fluoxetine group and tianeptine group,and the difference was statistically significant(F=33.343,P=0.000).The horizontal squares crossed,erection times,modification times and percentage of sacchar-consumption between the four groups showed no statistically significant differences before model establishment(P>0.05),but showed statistically significant differences after model establishment(P<0.05).Conclusion The expression of Cytochrome C in hippocampus of chronic stress depression rats decreases and can be reversed by some antidepressants through inhibiting the release of Cytochrome C of mitochondria in neuron,thus reducing neuronal apoptosis caused by stress.
