人脐带间充质干细胞移植对缺血性脑卒中模型大鼠行为学影响的研究

【摘要】 目的:观察人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,hUCMSCs)移植对缺血性脑卒中模型大鼠行为的影响。方法:制作大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)局灶性脑缺血大鼠模型,随机分为移植组和对照组,2 d后将体外培养的第3代hUCMSCs经尾静脉移植入移植组大鼠体内,对照组大鼠注入等量生理盐水。用Longa评分、转棒实验及Morris水迷宫等行为学检测方法,评价细胞移植后模型大鼠行为学改善情况。结果:(1)自细胞移植后第7天起,移植组Longa评分优于对照组(P<0.05);(2)细胞移植20 d后,移植组大鼠在转棒上停留时间与对照组相比明显延长(P<0.05);(3)细胞移植30 d后,移植组大鼠逃避潜伏期短于对照组(P<0.05)。结论:hUCMSCs可促进MCAO局灶性脑缺血模型大鼠神经功能的恢复。

【Abstract】 Objective: To observe the effect of human umbilical cord mesenchymal stem cells(hUCMSCs) transplantation on the functional behavior recovery of focal ischemic rat models.Methods: The rats were used to establish the middle cerebral artery occlusion(MCAO) model and divided randomly into control group and transplantation group.Two days later,the passage-3 hUCMSCs(3×106) were transplanted into the model animals via tail vein.The rats in control group were given the same dose of 0.9% saline.The functional behaviors were assessed by using Longa score,rotarod test and Morris water maze test.Results:(1)Since the 7th day after transplantation,Longa score of animals in transplantation group was markedly more than that in the control group(P<0.05).(2)At the 20th day after transplantation,the maintanence time of the transplantation group in rotarod test was significantly prolonged compared with the control group(P<0.05).(3)The Morris water amaze test showed that the escape latency of the transplantation group were shorter than that of the control group(P<0.05).Conclusion: hUCMSCs transplantation could promote neurological functional recovery of the MCAO model rats.
