慢性应激性刺激对大鼠行为的影响——抑郁症模型的建立

【摘要】 目的:以慢性应激性刺激(chronic mild stress,CMS)制作抑郁症动物模型,并以多种方式检测模型鼠的行为学变化,以期获得理想的动物模型。方法:对SD大鼠给予不可预知的慢性应激性刺激,包括:日夜颠倒;笼子弄脏;将笼子倾斜45°;限制它的活动;夜间不给予充足的食物;停止或减少对其水的供应等。每项刺激持续时间≥20 min,每2次刺激间隔时间≥16 h,为时5周。在刺激结束后,立即进行行为学测试,包括悬尾实验(tail suspension test,TST);强迫游泳实验(forced swim test,FST);蔗糖饮水实验(sucrose preference test,SPT)。结果:经过CMS处理的大鼠在悬尾试验中的不动时间明显大于未经过CMS实验处理的大鼠的不动时间(P<0.05)。在强迫游泳实验中,经过CMS处理的大鼠不动次数大于未经CMS实验的大鼠的次数,游泳的次数明显少于未经CMS实验处理的大鼠的游泳次数。未经CMS实验的大鼠蔗糖摄入百分比为(72.10±5.24)%,而经过CMS实验大鼠的摄入蔗糖水百分比为(52.98±11.14)%,明显低于对照组(P<0.05)。结论:不可预知的缓慢刺激长时间施加到啮齿类动物,可以模拟出类似抑郁症的相关症状。

【Abstract】 Objective: To generate a successful animal model of depression by exerting chronic mild stress(CMS) to rats followed by multiple behavior tests.Methods: The SD rats were stimulated with different stresses including reversion of day and night,dirty cage,45-degree incline of cage,movement limitation,shortage of food and water,and so on.The animals receive physical or mental torment for 5-week and each of stimulations lasts at least 20 min.The interval between two stimulates was 16 hours or more.When the stimulations were totally finished,animals were given a series of behavior tests including tail suspension test(TST),forced swim test(FST) and sucrose preference test(SPT).Results: TST results showed that the animals receiving CMS had significantly longer immobility time than those not receiving CMS(P<0.05).More times of motionless and less swimming were observed in the animals receiving FST than those in control group.SPT results indicated that percentage of sucrose intake in the CMS-treated animals(72.10 ±5.24)% was markedly more than those in untreated group(52.98±11.14)%(P<0.05).Conclusion: Depression-like symptoms can be elicited by CMS treatment which is suitable for generation of depression animal models.
