【摘要】 目的:本实验观察离焦性近视眼豚鼠中整合素和胶原蛋白在巩膜重塑中的作用。方法:-7D的凹透镜离焦14天建立豚鼠光学离焦性近视眼模型,RT-PCR法和免疫组化染色法检测各实验组后部巩膜中I型胶原α1(1)链、整合素α2、β1及ILK的mRNA及蛋白表达。结果:Ⅰ型胶原α1(1)链、整合素α2、β1和ILK在离焦组和恢复组的实验眼的mRNA水平及蛋白表达均减弱,与自身对照眼比较差异有显著统计学意义(P<0.01),去除离焦恢复2d后,上述指标表达回升,但与对照眼相比有统计学意义(P<0.05)。结论:整合素α2、β1、Ⅰ型胶原、LIK参与实验性近视眼巩膜重塑过程,是其早期调节因子。更多还原

【Abstract】 Objective: To observe the expression of the mRNA and protein level expression of integrin β1,α2,ILK in the defocus myopia sclera of guinea pigs,which may take part in remodeling process of the experimental myopia sclera.Methods: After 14 days optical defocus by means of-7D concave lens and establish the concave lens induced defocus myopia model of guinea pig,and the polymerase chain reaction(RT-PCR) and immunohistochemical staining to measure Ⅰ type collagenα1(1) chain,integrin α2,β1 and the level expression of the ILK of mRNA and protein of posterior sclera in each group.Results: The mRNA and protein level expression of I type collagen α1(1) chain,integrin β1,integrin α2 and ILK in the experimental eyes of the defocus groups and the restoration group is dropped.The differences with self comparison eyes have significant statistical significance(P < 0.01).After two days of recovery,compared with experimental eyes in the defocus group,they show that mRNA level expression of integrin type Ⅰ of collagen α1(1) chain,integrin β1,integrin α2 and ILK is increased.The difference has statistical significance(P <0.05).Conclusion: Integrin β1,integrin α2,ILK are early regulatory factors in the scleral remodeling process,which take part in remodeling process of the experimental myopia sclera.更多还原
