IL-13基因+2044G→A多态性与儿童支气管哮喘关联性研究
Association of IL-13 gene+2044G→A Polymorphism with Bronchial Asthma in Children
【摘要】 目的探讨IL-13基因+2044G→A多态性与潍坊地区汉族人群儿童支气管哮喘发病的关系。方法采用PCRRFLP方法,对90例支气管哮喘患儿(哮喘组)和82例健康儿童(对照组)IL-13基因+2044G→A多态性进行检测,计算基因型和等位基因频率,采用X2检验进行组间比较。结果两组人群IL-13基因十2044G→A多态性分布具有显著差异,GG,GA,AA基因型频率哮喘组分别为37.8%,54.4%和7.8%,对照组分别为68.3%,28.0%,3.7%(X~2=16.0,P<0.01);G等位基因和A等位基因频率哮喘组为65.0%和35.0%,对照组为82.3%和17.7%(X~2=13.14,P<0.01);哮喘组AA基因型和A等位基因频率明显高于对照组。与GG基因型和G等位基因比较,携带AA基因型和A等位基因的儿童发生支气管哮喘的相对风险明显增加,OR值分别为3.84(95%CI:3.83~14.70)(P<0.05)和2.51(95%CI:1.91~6.60)(P<0.01)。结论 IL-13基因+2044G→A多态性与儿童支气管哮喘的发病具有关联性,A等位基因可能是易感基因。
【Abstract】 Objective To confirm the role of IL-13 gene + 2044G→A polymorphism in children with bronchial asthma from the Han ethnic group in a weifang.Methods The IL-13 gene + 2044G→A polymorphism was determined by PCR-RFI.P in 90 patients with bronchial asthma(asthma group) and 82 normal children) control group).x~2 test was used for comparison between 2 groups after Calculation of genotype and allele frequency.Results The IL-13 gene +2044G→A polymorphism showed significant differences between the two groups.The frequencies of GG,GA and AA genotypes of IL-13 gene + 2044G→A polymorphism were,respectively,37.8%,54.4%and 7.8%in group of asthma,68.3%,28.0%and 3.7%in group of control(x~2= 16.0,P<0.01).The G and A allele frequencies of IL-13 gene were,respectively,65.0%and 35.0%in group of asthma,82.3%,and 17.7%in group of control(x~2= 13.14,P <0.01).The frequencies of AA genotype and A allele for IL-13 gene +2044G→A revealed significantly higher in asthma group than in control group.The OR of AA genotype and A allele were 3.84(P <0.05) and 2.51(P < 0.01).Conclusion In the present study,it confirmed the association of IL-13 gene +2044G→A polymorphism with bronchial asthma in children,and found A allele may be one of the susceptibility factors for bronchial asthma.
