蜂胶黄酮对环磷酰胺诱导HL7702细胞损伤的保护作用

【摘要】 目的探讨蜂胶黄酮(EPF)对环磷酰胺(CTX)诱导HL7702细胞凋亡的保护作用。方法将HL7702细胞按一定密度分别接种于96孔板和24孔板,培养24 h后,向各孔中分别加入CTX(4 ng/L)、CTX(4 ng/L)+EPF(20、30、40 ng/L),继续培养24 h,进行WST-1、AO/EB双染、Annexin-V-FITC/PI流式细胞染色。采用小鼠随机分组,试验组每天给予EPF 100、200、400 mg/mL灌胃,0.5 mL/支,连续给药10 d,同时注射CTX0.15 g/kg,共3 d。取血清检测过氧化氢酶(CAT)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、谷丙转氨酶(GPT)、谷草转氨酶(GOT)等活性及尿素氮(BUN)含量。结果 EPF与CTX共处理HL7702细胞生长抑制明显改善,当EPF浓度达到40 ng/mL时,促进生长作用呈现正比例曲线特征。试验组凋亡细胞明显减少,绝大多数细胞形态饱满,几乎没有死亡细胞。CTX单独处理的HL7702细胞组24 h晚期凋亡细胞占30.2%,CTX与EPF共处理细胞组凋亡细胞分别占19.2%、13.1%、7.1%。结论 EPF对CTX诱导的HL7702凋亡作用具有拮抗作用,可缓解CTX引起的毒性。

【Abstract】 Objective To investigate the protective effect of extract of propolis flavone(EPF) on HL7702 cells apoptosis induced by CTX.Methods HL7702 cells were cultured in 96 well plates and 24 well plates for 24 h,then CTX(4 ng/mL)and CTX(4 ng/mL)+EPF(20,30,40 ng/mL)were added.After 24 h,WST-1,AO/EB double staining and Annexin-V-FTTC/PI staining were performed.The mice were divided randomly into 5 groups,experimental group was given EPF(100,200,400 mg/mL,0.5 mL/d)for 10 d and CTX 0.15 g/kg for 3 d.Serum Catalase(CAT),lactate dehydrogenase(LDH),creatinekinase(CK),glutamic-pyruvic transaminase(GPT),glutamic-oxaloacetic transaminase(GOT)and blood urea nitrogen(BUN)were observed.Results EPF reduced the inhibitory effects of CTX on HL7702 cell growth,and it promoted the growth and illustrated direct proportion curve character when the concentration up to 40 ng/mL.The amount of apoptosis decreased in experiment group,and the morphology of cell was satiation with almost no dead cell.The percentage of later apoptotic cells of CTX group accounted for 30.2% in 24 h,while the percentage of later apoptotic cells of CTX and EPF group was 19.2%,13.1% and 7.1% respectively.Conclusion EPF can antagonize the apoptosis of HL7702 cell and reduce the toxicity induced by CTX.
