大鼠环孢霉素A中毒性肾损伤模型的探讨

【摘要】 目的:大鼠环孢霉素A中毒性肾损伤模型的探讨。方法:实验大鼠随机分为对照组,不同浓度的环孢霉素A给药组,分别给予橄榄油灌胃,不同浓度的环孢霉素A灌胃,连续4w后,测动物体重,24 h尿量,尿蛋白,取血测血肌酐、尿素氮,取肾组织作病理观察并行肾小管损伤评分,测组织超氧化物歧化酶和丙二醛含量。结果:与对照组比,25 mg/kg/d环孢霉素灌胃组肾组织结构变化明显,但功能改变不明显,50 mg/kg/d环孢霉素灌胃组肾脏形态结构及功能受损均明显,尿蛋白含量增多,血肌酐、尿素氮明显增高(P<0.05);肾组织超氧化物歧化酶含量减低而氧化产物丙二醛MDA含量明显增高(P<0.05)。结论:持续4周50 mg/kg/d剂量环孢霉素A灌胃可复制典型的环孢霉素中毒性肾损伤模型。

【Abstract】 Objective: To explore the establishment of nephrotoxicity model by Cyclosporin A(CsA).Methods:Rats in 3 groups were treated with olive oil(control) and CsA of two different doses respectively.After 4 weeks,nephrotoxicity was assessed by the examination of body weight, serum creatinine, blood urea nitrogen, activity of antioxitant and histopathological changes. The injury of renal tubules was scored. Results: Both histopathological and functional damages were significant in CsA 50mg/kg/d group; the functional change was less conspicuous in CsA 25mg/kg/d group. Conclusion: The result indicates that a nephrotoxicity animal model can be replicated by4 weeks intragastric administration of cyclosporine at the concentration of 50 mg/kg/d.
