不同浓度的高锶矿泉水对人肝细胞增殖及功能的影响

【摘要】 目的:观察不同浓度的高锶矿泉水对LO2细胞(人肝细胞株)增殖和功能的影响。方法:LO2细胞分为2mg/L、4mg/L、6mg/L、8mg/L锶矿泉水组(SMW)和双蒸水组(DDW),以5×104/ml的浓度接种于培养板中,24h后,换成条件培养基培养至7d。分别于2d、3d、5d和7d后行MTT法观察细胞的增殖情况。培养5d的细胞分别用比色法测培养基中总胆红素(TB)的含量,和用赖氏法测细胞内谷丙转氨酶(ALT)和谷草转氨酶(AST)的活性。结果:2mg/L SMW细胞增殖、胆红素摄取量和转氨酶活性无显著变化(P>0.05);4mg/L和6mg/L SMW组细胞提前出现增殖峰,并较长时间地保持旺盛的增殖状态(P<0.05);细胞摄取胆红素的能力增强(P<0.05),AST活性无显著变化(P>0.05),但ALT的活性降低(P<0.05);8mg/L SMW组细胞增殖速度减慢、摄取胆红素量增多(P<0.05),但细胞内ALT和AST的活性均降低(P<0.05)。结论:4~6mg/L浓度范围的天然高锶矿泉水对肝细胞的增殖和胆红素代谢有益,能降低转氨酶活性;但超过8mg/L浓度时可对肝细胞造成一定损害。

【Abstract】 Objective: To investigate the effect of different concentration of strontium mineral water(SMW) on proliferation、bilirubin metabolism and transaminase activity of human hepatocytes(LO2 cells).Methods: Human hepatocytes were cultured in vitro.After cultured in a normal Dulbecco’s Modified Eagle’s Medium(high glucose) for 24 hours,the cells were cultured in conditioned medium containing 2,4,6,8 mg/L strontium mineral waters and double distilled water(DDW).The proliferation of LO2 cells were observed by MTT assay at 3,5,7d.The level of total bilirubin of mediums were measured by sodium、benzoate-caffeine colorimetric when cells cultured at 5d.Alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activity in cells were determined by Reitman’s.Results: 2 mg/L SMW did not affect proliferation bilirubin metabolism and transaminase activity of LO2 cells.The cell proliferation peak in 4mg/L and 6mg/L SMW groups came earlier than in DDW group,and kept vigorous for a long time(P<0.05).The bilirubin intake of cells in 4mg/L and 6mg/L SMW increased when cultured at 5d.While activity of ALT decreased.LO2 cells in 8mg/L SMW grew slower than in DDW(P<0.05),and transaminase(ALT and AST) activity decreased and the bilirubin intake increased(P<0.05).Conclusion: The Strontium mineral water is good to the cell proliferation and bilirubin metabolism of hepatocytes at 4mg/L and 6mg/L.But it will damage hepatocytes over 8mg/L.
