【摘要】 目的探讨金属基质蛋白酶9(MMP-9)在结肠癌细胞膜型MICA(mMICA)脱落为可溶型MICA(sMICA)过程中的作用。方法 HT-29细胞转染MMP-9siRNA质粒沉默48小时后,检测MMP-9、MICAmRNA表达以及MMP-9、mMICA及sMICA蛋白生成情况。结果 MMP-9siRNA质粒转染HT-29细胞后显著降低MMP-9mRNA,不影响MICAmRNA;细胞膜表面mMICA蛋白显著增加同时sMICA蛋白生成明显减少。结论抑制MMP-9表达,可减少结肠癌细胞mMICA蛋白降解,同时下调sMICA蛋白生成。更多还原

【Abstract】 Objective To investigate the effect of MMP-9 in the process of mMICA shedding to sMICA in human colon cancer cells.Methods MMP-9 siRNA plasmid was transfected into HT-29 cells,the expressions of MMP-9 and MICA mRNA were detected through qPCR,he proteins of MMP9,sMICA and mMICA were detected by ELISA.Results Transfection of MMP-9 siRNA into HT-29 resulted in a significant decrease in the levels of MMP-9 mRNA and protein,decreased the shedding of sMICA form mMICA,and upgraded the produce of protein of mMICA,however,it didn’t affect the expression of mRNA of MICA.Conclusion Inhibiting the expression of MMP-9 can decrease the porduction of sMICA as well as increase mMICA.更多还原
