美洲黑杨与胡杨杂交及其杂种子代亲子鉴定

【摘要】 以美洲黑杨为母本、胡杨为父本,对克服胡杨与杨属其它派树种杂交障碍技术进行研究,结果表明:美洲黑杨花粉提取液处理柱头能有效克服美洲黑杨与胡杨杂交受精前障碍,而幼胚培养技术能克服其受精后障碍。同时,运用形态学标记、SSR分子标记技术对杂交子代及其亲本进行鉴定分析,形态学标记表明,杂种子代主要偏向于中间型,偏父本型、母本型较少;SSR分子标记也表明,杂交子代均显示出双亲间的杂种类型谱带,从而可以在分子水平上证明美洲黑杨与胡杨间杂交的可交配性。

【Abstract】 The research on technology of overcoming the crossing barrier between Populus euphratica and other Populus species was conducted by using P. deltoides as maternal parent and P. euphratica as paternal parent. The results showed that pre-fertilization barriers could effectively be overcome by using P. deltoides pollen extract to treat stigma and post-fertilization barriers could be overcome by immature embryo culture technique. At the same time, the hybrids and their parents were identified using morphological marker and SSR molecular markers. Morphological test showed that the vast majority of hybrids offspring were intermediate type and little were partial parent type; SSR test showed that all hybrids have the heterozygous bands caused by the recombination between the both parents. The molecular evidence indicated the cross ability between P.deltoides and P.euphratica.
