荧光素-Fe(Ⅲ)配合物与BSA的相互作用的光谱学研究
Spectroscopic Studies on Interaction of BSA Molecules and Fluorescein-DA-Fe(Ⅲ) Complex
【摘要】 双[N,N-双(羧甲基)氨甲基]荧光素(荧光素-甲胺基二羧酸)是一种含有三环平面结构的荧光素衍生物.研究采用荧光素-甲胺基二羧酸为配体,采用铁离子(Fe(Ⅲ))为中心金属离子,通过直接反应的合成方法,在水溶液中合成了荧光素-甲胺基二羧酸-Fe(Ⅲ)配合物.并采用紫外-可见(UV-vis)光谱和荧光(FL)光谱来研究荧光素-甲胺基二羧酸-Fe(Ⅲ)配合物与牛血清白蛋白(BSA)的相互作用.相应的研究数据和计算结果表明荧光素-甲胺基二羧酸-Fe(Ⅲ)配合物对BSA分子荧光的猝灭机理是静态猝灭过程.同时采用三维荧光光谱和三维等高线剖面图谱进一步考察了荧光素-甲胺基二羧酸-Fe(Ⅲ)配合物存在时BSA分子的荧光猝灭过程和混合前后的构象变化.通过同步荧光光谱研究了荧光素-甲胺基二羧酸-Fe(Ⅲ)配合物与BSA分子相互作用的结合方式和结合位点.研究为荧光素及其衍生物在医疗领域中的进一步应用奠定了基础.
【Abstract】 (Bis[N,N-bis(carboxymethyl) aminomethyl] fluorescein((2-(6-Hydroxy-3-oxo-(3H)-xanthen-9-yl) benzoic acid))(Fluorescein-DA) is a kind of fluorescein derivant with a tricyclic plane structure.In this work,Fluorescein-DA was adopted as the ligand and iron ion(Fe(Ⅲ) was selected as the centre metal ion.And then,Bis[N,N-bis(carboxymethyl) aminomethyl] fluorescein-Ferrous(Ⅲ)(Fluorescein-DA-Fe(Ⅲ)) complex was synthesized through direct reaction in aqueous solution.The interaction between Fluorescein-DA-Fe(Ⅲ)) complex and bovine serum albumin(BSA) was studied by using fluorospectrometry and UV-vis spectrophotometry.As judged from the experimental data and calculative results,the fluorescence quenching of BSA caused by Fluorescein-DA-Fe(Ⅲ) was attributed to static quenching process.Three-dimensional fluorescence spectra and three-dimensional fluorescence contour profile spectra were mentioned to determine the fluorescence quenching and the conformation change of BSA in the absence and presence of Fluorescein-DA-Fe(Ⅲ).The binding pattern and binding site of Fluorescein-DA-Fe(Ⅲ) and BSA molecules were determined by the synchronous fluorescence spectroscopy.Perhaps,this paper may offer some important subjects for broadening the application of fluorescein and its complexes in medical treatment.
