钙腔蛋白rs339097基因多态性与华法林稳定剂量的关系

【摘要】 目的:探讨钙腔蛋白(CALU)rs339097基因型在中国汉族人群中的分布情况及其基因多态性与瓣膜置换术后华法林稳定剂量的关系。方法:采用Snapshot技术检测226例研究对象CALU rs339097位点基因型,计算其基因型及等位基因频率。对其中176例瓣膜置换术患者,分析CALU rs339097基因多态性与华法林稳定剂量的相关性。结果:226例研究对象中,基因型TT、CT、CC分别为221例(97.8%)、4例(1.8%)和1例(0.4%),T和C等位基因分别为446次(98.7%)和6次(1.3%)。176例瓣膜置换术患者中,CC基因型未检出,TT基因型检出172例,CT基因型检出4例;在达到稳定抗凝状态时,TT基因型和CT基因型华法林日维持剂量[(3.19±0.86)mg/d∶(4.50±0.61)mg/d]差异有统计学意义(P=0.003)。结论:中国汉族人群CALUrs339097基因多态性是瓣膜置换术后华法林个体剂量差异的影响因素。

【Abstract】 Objective:To investigate the genotypes distribution of calumenin(CALU) rs339097 in Chinese Han population,and the relationship between CALU rs339097 polymorphism and the stable dose of warfarin in the patients after valve replacement.Method:The genotypes and allele frequencies of 226 cases were detected by Snapshot technology.The 176 patients after valve replacement were selected to investigate the relationship between gene polymorphism and the stable dose of warfarin.Result:In total of 226 cases,the number of TT,CT,CC genotypes were 221(97.8%),4(1.8%) and 1(0.4%).The frequencies of T and C allele were 98.7% and 1.3%.In 176 patients after the operation,CC genotype was not detected,TT genotype was detected in 172 patients,and CT genotype was detected in 4 patients.The maintenance stable dose of warfarin with TT and CT were(3.19±0.86)mg/d,(4.50±0.61)mg/d,P=0.003.Conclusion:The CALU rs339097 polymorphism in Chinese Han population is an important factor affecting individual warfarin maintenance dose in the patients after the valve replacement surgery.
