枸杞多糖和原花青素对小鼠成釉细胞DNA损伤作用的研究

【摘要】 目的:探讨枸杞多糖和原花青素对小鼠成釉细胞DNA损伤的保护作用。方法:小鼠成釉细胞以100.00、200.00、400.00 mg/L枸杞多糖作为枸杞多糖作用组,以10.00、25.00、50.00 mg/L原花青素作为原花青素作用组。细胞染毒24 h后,采用单细胞凝胶电泳技术(SCGE)检测DNA单链断裂情况。结果:与对照组比较,100.00、200.00和400.00 mg/L枸杞多糖作用组小鼠成釉细胞尾长、Olive尾距和尾长/头长值均明显下降,差异具有统计学意义(P<0.05)。100.00、400.00 mg/L枸杞多糖作用组尾DNA%较对照组明显下降(P<0.05)。与对照组比较,10.00、25.00、50.00 mg/L原花青素作用组小鼠成釉细胞DNA损伤的尾长、Olive尾矩、尾DNA%、尾长/头长值均较对照组均显著性下降,差异具有统计学意义(P<0.05)。以Olive尾矩为观察指标,100.00 mg/L枸杞多糖作用组的Olive尾矩明显低于200.00 mg/L和400.00 mg/L枸杞多糖作用组(P<0.05),10.00 mg/L原花青素作用组的Olive尾矩明显低于25.00 mg/L和50.00 mg/L原花青素作用组(P<0.05)。结论:100.00~400.00 mg/L枸杞多糖和10.00~50.00 mg/L原花青素对小鼠成釉细胞DNA损伤具有较好的保护作用,以100.00 mg/L枸杞多糖、10.00 mg/L原花青素的保护作用相对较好。

【Abstract】 Objective:To investigate the protective effects of Lycium barbarum polysaccharides(LBP)and Procyanidins(PC) on DNA damage in mouse ameloblasts. Method:The mouse ameloblasts were treated with LBP at the doses of 100.00,200.00 and 400.00 mg/L,and PC at the doses of 10.00,25.00 and 50.00 mg/L respectively. After exposed 24 h, the single cell gel electrophoresis assay was used to measure the DNA damage in mouse ameloblasts. Result:The tail length, olive tail moment and tail/head length ratio of LBP group at the doses of 100.00,200.00 and 400.00 mg/L were lower than those of the control group in mouse ameloblasts(P <0.05). LBP at the doses of 100.00 and 400.00 mg/L could decrease DNA % compared with contro(lP <0.05). PC at the doses of 10.00,25.00 and 50.00 mg/L decreased the tail length, olive tail moment,DNA % and tail/head length ratio in mouse ameloblasts compared with control group(P <0.05). Selected olive tail moment as effective indicator,the protective effects of LBP at the dose of 100.00 mg/L were better than those of 200.00 and 400.00 mg/L LBP,and also 10.00 mg/L PC better than those of 25.00 and 50.00 mg/L PC. Conclusion:It was suggested that Lycium barbarum polysaccharides at the dose of 100.00~400.00 mg/L and Procyanidins at the dose of 10.00~50.00 mg/L could inhibit DNA damage in mouse ameloblasts respectively,100.00 mg/L Lycium barbarum polysaccharides and 10.00 mg/L Procyanidins have relative better protective effect.
