纳米荧光碳点与细菌生物相容性的初步研究
Preliminary investigation on the biocompatibility of fluorescent carbon dots with Escherichia coli and Staphylococcus aureus
【摘要】 目的用大肠埃希菌(E.coli)和金黄色葡萄球菌(S.aureus)研究新型免疫标记物——纳米荧光碳点(CDs)对此两种细菌的生物相容性。方法以洽草为材料合成CDs,通过透射电镜(TEM)﹑荧光光谱仪和傅立叶红外分析仪研究合成的CDs的特征。借助比色计和扫描电镜(SEM)观察这两种细菌分别与CDs共培养后的生长曲线,及CDs对细菌形态的影响。用傅立叶变换衰减全反式红外分析(ATR-FTIR)和荧光光谱仪分析此两种菌对CDs的直接效应。结果合成得到形态较均一的CDs。与细菌共培养的CDs对E.coli和S.aureus的生长无明显影响,但可影响细菌的形态;细菌能够黏附或吞噬一定量的CDs,并可致CDs的荧光衰减。结论 CDs对E.coli和S.aureus有一定的生物相容性,但有细胞毒性;而此两种菌对CDs荧光有非特异性猝灭作用。
【Abstract】 Objective To investigate the biocompatibility of fluorescent carbon dots(CDs),a new type of immune-labelling material,with Escherichia coli(E.coli) and Staphylococcus aureus(S.aureus).Methods CDs was synthesized with Koeloriacristata,and its characteristics was evaluated with transmission electron microscopy(TEM),fluorescence spectroscopy and Fourier infrared analyzer.The growth curves of E.coli and S.aureus cocultured with CDs respectively,and the effects of CDs on their morphology were observed by densitometer and scanning electron microscopy(SEM).While,the direct effects of E.coli and S.aureus on CDs were analyzed with Fourier transform attenuated total reflection infrared analysis(ATR-FTIR) and fluorescence spectroscopy.Results The CDs with a uniform morphological characteristic were synthesized successfully,and they had no obvious effect on the growth of E.coli and S.aureus,but influenced their morphology.However,E.coli and S.aureus could adsorb or phagocytize a certain amount of CDs and quench the fluorescence of CDs.Conclusion The synthesized CDs had good biocompatibility with E.coli and S.aureus,but also displayed some cytotoxicity.Meanwhile,E.coli and S.aureus could nonspecifically quench the fluorescence of CDs.
