【摘要】 目的检测白细胞介素17(IL-17)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)在原发性干燥综合征患者唇腺组织中的表达,分析其意义。方法采用免疫组织化学链霉菌抗生素蛋白-过氧化物酶连接法(SP)法检测IL-17、IL-6、TNF-α在30例原发性干燥综合征、10例对照组唇腺组织在IL-17的表达,测得其光密度值。结果 IL-17、IL-6、TNF-α在原发性干燥综合征唇腺的表达明显高于正常组,分别为0.078±0.012vs 0.210±0.077;0.079±0.014vs 0.245±0.083;0.081±0.007vs 0.251±0.073(P<0.01)。30例pSS患者中,按照淋巴细胞浸润程度分级,Ⅰ级5例,Ⅱ级7例,Ⅲ级12例,Ⅳ级6例。IL-17在Ⅰ、Ⅱ、Ⅲ、Ⅳ级间的表达分别为0.094±0.029,0.171±0.052,0.240±0.030,0.293±0.050。IL-6在各级别间的表达分别为0.120±0.06611,0.2443±0.070,0.281±0.064,0.278±0.050,TNF-α在各级别间的表达分别为0.138±0.049,0.241±0.067,0.283±0.047,0.290±0.043。经方差分析各级别间IL-17、IL-6、TNF-α表达差异均有统计学意义(P<0.05)。且IL-17、IL-6、TNF-α的阳性表达程度与淋巴细胞浸润程度呈正相关(r=0.814、0.469、0.591,均P<0.01)。结论 IL-17、IL-6、TNF-α均参与了pSS的局部炎症反应过程,且与淋巴细胞浸润程度呈正相关。提示Th17细胞及其细胞因子在pSS的发病过程中起着重要作用。更多还原

【Abstract】 Objective The purpose of this study is to investigate the expression and significance of interleukin17(IL-17),interleukin 6(IL-6)and tumor necrosis factorα(TNF-α)in the labial salivary glands of patients with primary Sjogren syndrome(pSS).Methods The expression of IL-17,IL-6and TNF-α were detected by using immunohistochemistry SP method in 30patients with pSS and 10control subjects,the optical density values were measured in the same paraffin section.Results The average optical density of IL-17,IL-6,TNF-αin pSS group revealed significantly higher values in comparison to those in control group,respectively 0.078±0.012vs 0.210±0.077;0.079±0.014vs 0.245±0.083;0.081±0.007vs 0.251±0.073(P <0.01).In 30pSS cases,there were 5gradeⅠ,7gradeⅡ,12gradeⅢand 6gradeⅣ,according to the degree of lymphocyte infiltration classification.The positive expression of IL-17inⅠ,Ⅱ,Ⅲ,Ⅳ grade were 0.094±0.029,0.171±0.052,0.240±0.030,0.293±0.050,respectively,the positive expression of IL-6,0.120±0.066,0.244±0.070,0.281±0.064,0.278±0.050,the positive expression of TNF-α0.138±0.049,0.241±0.067,0.283±0.046,0.290±0.043.Through variance analysis,the expression of IL-17,IL-6,TNF-αhad statistic significance between different grades(P <0.05).There were positive correlation between positive expression of IL-17,IL-6and TNF-αand the grades of lymphocyte infiltration(r=0.814,0.469,0.591,all P <0.01).Conclusion IL-17,IL-6and TNF-αparticipated in the local inflammatory reaction of labial salivary glands in pSS patients.There are correlation between the expression of three cytokines and lymphocyte infiltration.Th17cytokines may play an important role in the pathogenesis of pSS.更多还原
