化湿通瘀补气法对延缓家兔动脉粥样硬化斑块形成的实验研究

【摘要】 目的观察化湿通瘀补气法延缓高脂饮食兔动脉粥样硬化(AS)斑块形成的作用。方法纯种新西兰雄性大白兔(4个月月龄)18只作为实验对象,常规喂养1周后随机分为3组,每组6只。改为高脂饮食(1%胆固醇+6%蛋黄粉+8%猪油+85%基础饲料)喂养,对照组不用药,中药干预组用荷叶、泽泻、虎杖等中药,西药干预组用阿司匹林+阿托伐他汀+普罗布考。各组3个月后分别抽取耳缘静脉血,测定总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、基质金属蛋白酶9(MMP-9)、丙二醛(MDA)、氧化低密度脂蛋白(ox-LDL)值,然后处死动物,取主动脉组织作苏木精-伊红染色切片及免疫组化染色,观察主动脉病理组织学变化,测定各组主动脉内膜及内膜/中膜厚度。结果中药干预组和西药干预组的TC、TG和LDL-C均比对照组明显降低(P<0.01),TC(2.39±0.15)mmol/L,(2.08±0.12)mmol/L vs(13.02±0.20)mmol/L;TG(2.58±0.16)mmol/L,(1.90±0.08)mmol/L vs(11.08±0.19)mmol/L;西药干预组还低于中药干预组(P<0.01),LDL-C(1.05±0.24)mmol/L,(0.85±0.09)mmol/L vs(10.13±0.20)mmol/L(P<0.01);MDA、ox-LDL、MMP-9水平较对照组均有不同程度降低(P<0.01)。中药干预组和西药干预组主动脉内膜厚度较对照组明显降低,分别为(12.28±1.28)μm、(5.20±0.81)μm vs(40.06±3.94)μm(P<0.01),而西药干预组比中药干预组降低明显。结论化湿通瘀补气法能延缓兔动脉内斑块形成,降低LDLC,为中医药治疗及动脉粥样硬化提供新的研究理论依据。

【Abstract】 Objective The study was to observe the delaying of atherosclerotic(AS)plaque formation in rabbit fed with high lipid diet by Huashi Tongyu Buqi Method.Methods Pure bred 18New Zealand male white rabbits(4months old)were experimental subjects.After normal feeding for one week,the rats were changed to high lipid diet(1% cholesterol+ 6% yolk powder + 8% lard +85% basic fodder),and they were randomly divided into three groups:six rabbits in control group(without using any drugs),six rabbits in Chinese medicine group(Heye,Zexie,Huzhang,etc)and six rabbits in western medicine group(aspirin+ atorvastatin+ probucol)and observed for three months.After three months,auricular venous blood of each group was drawn and total cholesterol(TC),triglycerides(TG),low-density lipoprotein-cholesterol(LDL-C),serum matrix metalloprotinase-9(MMP-9),malondialdehyde(MDA)and ox-LDL were measured,and then rabbits were killed to get aorta.Hematoxylin-eosinstaining and immunohistochemistry staining were used to observe the histopathological changes of aorta,atherosclerotic plaque area,intima and vessel wall thickness.Results TC,TG and LDL-C of Chinese medicine group and western medicine group were significantly lower than those of control group(P<0.01),respectively TC(2.39±0.15)mmol/L,(2.08±0.12)mmol/L vs(13.02±0.20)mmol/L;TG(2.58±0.16)mmol/L,(1.90±0.08)mmol/L vs(11.08±0.19)mmol/L;LDL-C(1.05±0.24)mmol/L,(0.85±0.09)mmol/L vs(10.13±0.20)mmol/L,TC and TG of western medicine group was much lower than that of Chinese medicine group(P<0.01);MDA,MMP-9and ox-LDL level as compared with control were decreased by different extents(P <0.01).Aortic intima thickness of Chinese medicine group and western medicine group decreased significantly as compared with that of control group,(12.28±1.28)μm,(5.20±0.81)μm vs(40.06±3.94)μm(P <0.01),while aortic intima thickness of western medicine group showed more decrease.Conclusion Huashi Tongyu Buqi method delays the atherosclerotic plaque formation in rabbit,decreases blood LDL-C,provides new research theoretical evidence for Chinese medicine and atherosclerosis.
