应用超声心动图检测心肌梗死大鼠恢复期心脏结构与功能改变

【摘要】 目的探讨超声心动图检测心肌梗死大鼠恢复期心脏结构与功能的可行性、准确性及敏感性。方法 47只正常清洁级雄性Sprague-Dawley(SD)大鼠,随机分为心肌梗死组23只和假心肌梗死组24只,均常规麻醉开胸,心肌梗死组于左心耳2 mm处丝线结扎左冠状动脉前降支;假心肌梗死组仅穿线不结扎冠状动脉。造模8周后进行超声心动图检查,获得大鼠心脏形态学及功能学指标,然后处死大鼠对心脏行2,3,5-氯化三苯基四氮唑(TTC)染色,并与超声心动图检测结果进行关联性分析。结果心肌梗死组23只大鼠中,经超声心动图检出心肌梗死14只,经TTC染色提示大鼠存在心肌梗死者16只,故超声心动图检测心肌梗死大鼠的敏感性为87.5%(14/16),特异性为100%(7/7);超声心动图检测结果与心脏TTC染色具有良好地关联性(Carmer’s V=0.91且P<0.05)。结论超声心动图可用于准确评估心肌梗死大鼠恢复期心脏结构与功能的变化。

【Abstract】 Objective To evaluate the feasibility,accuracy and sensitivity of the echocardiography in the detection of cardiac structural and functional changes in rats myocardial infarction convalecence.Methods Forty-seven normal Sprague Dawley male rats wer enrolled in,and they were randomly divided into myocardial infarction(MI)group(23 rats)and the sham control group(24 rats).The left anterior descending artery was ligated at two millimeters below the bottom of the left appendage in MI group,while in the sham control group,only a surgical suture was passed through the bottom of the left anterior descending artery and without ligation.At the time of eight weeks after the operation,the cardiac morphological and functional parameters of all rats were obtained by echocardiography.Then all of rats were sacrifcied and the cardiac tissues were stained with 2,3,5 chlorinated-triphenyl-tetrazole(TTC).Results Fourteen cases were detected myocardial infarction using echocardiography in MI group,while 16 cases were verified myocardial infarction with TTC staining in the same group.The sensitivity of echocardiography in detecting rats myocardial infarction was 87.5% and the specificity was 100%.In addition,the correlation coefficient between the TTC staining and echocardiography for detecting myocardial infarction was 0.91(P < 0.05).Conclusion Echocardiography is an accurate inspection method in detecting and can be used to assess myocardial infarction in rats.
