金丝桃素对病毒性心肌炎小鼠慢性期的心肌保护作用

【摘要】 目的探讨金丝桃素(Hyp)对病毒性心肌炎小鼠慢性期心肌损害的影响。方法 60只健康雄性Balb/c小鼠腹腔注射柯萨奇病毒B3(CVB3)建立病毒性心肌炎心肌纤维化模型;另外10只注射Eagle’s MEM液作为对照,两个月后存活小鼠随机分为4组:模型组,Hyp大、小剂量组,卡托普利组;分别以Hyp及卡托普利每日灌胃给药一次,对照组及模型组给予同体积的生理盐水,30 d后,观察各组小鼠死亡率及心肌病理改变;检测血清白介素-2(IL-2)、血清可溶性白介素-2受体(sIL-2R)、血清肌酸肌酶(CK),以及细胞间黏附分子-1(ICAM-1)表达水平。结果与对照组相比,模型组小鼠死亡率显著增高,差异有统计学意义(P<0.05);模型组小鼠心肌细胞肿胀、扭曲,纤维化程度明显,ICAM-1表达,IL-2、sIL-2R水平及CK活性升高,差异均有统计学意义(P<0.01);Hyp或卡托普利组小鼠的死亡率均比模型组低,心肌病理损害减轻,ICAM-1表达及血清IL-2、sIL-2R水平及CK活性降低。结论 Hyp对病毒性心肌炎慢性期的心肌损害有保护作用,可能与抑制ICAM-1的表达、降低血清IL-2、sIL-2R水平有关。

【Abstract】 Objective To investigate the effect of hypericin on the myocardial injury in the chronic phase of coxsackievirus B3induced myocarditis in mice.Methods Sixty healthy male Balb/c mice were intraperitoneally inoculated with coxsackievirus B3 at fortnightly intervals to establish the model of chronic viral myocarditis.Ten mice were inoculated with EMEM solution and were used as controls.The mice surviving more than 60 days were randomly divided into four groups,model group,hypericin group with low or high dose and captopril group.Mice were intragastrically administered with captopril in captopril group or hypericin in hypericin group once a day.The mice in control group and model group were received equivalent volume of normal saline.After 30 days,mortality of mice in each group was recorded and myocardial histopathological changes were observed.The serum interleukin-2(IL2),soluble interleukin-2 receptor(sIL-2R),creatine kinase(CK) activity and the expression levels of intercellular adhesion molecule-1(ICAM-1) were detected.Results Compared with the control group,mortality of mice in model group was increased significantly(P<0.05).Myocardial cells of mice in model group showed obvious swelling,twist and fibrosis,and the expression level of ICAM-1 in myocardium,IL-2,sIL-2R levels and CK activity in serum of mice in model group were significantly higher than those of control group(P<0.01).Mortality of mice in hypericin group and captopril group were lower than that in model group.After the treatment with hypericin and captopril,pathological injury in myocardium was alleviated,and the expression of ICAM-1 in the myocardium and serum IL-2 level were decreased,while sIL-2R levels and activity of CK were decreased.Conclusion Hypericin may have a protective effect against myocardial injury in chronic phase of viral myocarditis by inhibition of the expression of ICAM-1,and reduction the levels of IL-2 and sIL-2R in serum.
