不同尺寸二氧化硅纳米颗粒体内分布与代谢研究
In vivo study of biodistribution and urinary excretion of silica nanoparticles with different size
【摘要】 基于二氧化硅荧光纳米颗粒(FSiNP)的荧光信号同步指示功能,通过实时、原位活体荧光成像技术,并结合离体器官成像、组织切片成像以及尿液荧光成像等方法,系统地研究了不同尺寸二氧化硅荧光纳米颗粒在裸鼠活体内的分布与代谢.活体成像结果表明纳米颗粒尺寸越小,血液循环时间越长,全身分布情况越明显,随着时间的延长纳米颗粒逐渐聚集到肝脏、膀胱等部位;通过离体器官成像和组织切片成像进一步证实了活体成像所观察到的结果.同时,肾脏组织切片以及尿液成像结果显示,颗粒越小越容易通过泌尿系统排出体外.研究结果为今后开展不同尺寸纳米颗粒材料在活体内的应用研究打下基础.
【Abstract】 The biodistribution and urinary excretion of fluorescent silica nanoparticles(FSiNPs) with different sizes were investigated in situ and real-time in mice using small animal imaging system.In addition,in vitro organ imaging,organization slice imaging and urine fluorescence imaging were also investigated.In vivo imaging results showed that the smaller the size of FSiNPs,the longer the blood circulation,and the more obvious in whole body distribution.Finally,these particles gradually accumulated in the liver and bladder.These results were well confirmed by in vitro organ imaging and organization slice imaging.At the same time,the kidney tissue sections and urine imaging results showed that the smaller the particle the more easily excreted through the urinary system.The results of this study would be helpful for future application of fluorescent silica nanoparticles with different sizes in vivo.
