【摘要】 目的:研究齐墩果酸(Oleanic acid,OA)对人白血病HL-60细胞PI3K、Akt、Cav-1表达的影响。方法:OA作用HL-60细胞后,采用透射电镜观察细胞形态,RT-PCR法检测HL-60细胞PI3K、Akt、Cav-1的mRNA表达。结果:OA作用于HL-60细胞后,细胞形态改变明显,出现凋亡小体,且PI3K、Akt、Cav-1的mRNA表达下调(P<0.01)。结论:OA可抑制HL-60细胞增殖,这可能与其下调HL-60细胞中PI3K、Akt、Cav-1的表达有关。更多还原

【Abstract】 Objective:To study the effect of oleanolic acid(OA) on the expression of pi3k、akt and cav-1 genes in leukaem HL-60 cells.Methods:In HL-60 cells,after being treated by OA,the morphological and structural changes of the cells were observed by transmission electro microscopy; the mRNA expressions of pi3k、akt and cav-1 were measured by RT-PCR.Results:After being treated by OA,the HL-60 cells had obvious morphological and structural changes under the transmission electro microscopy,and apoptosis body was observed as well.The mRNA expression of pi3k and akt were down regulated(P < 0.01),the mRNA expression of cav-1 was up regulated(P < 0.01).Conclusion:OA could inhibit the proliferation of HL-60 cells by downregulating the expression of PI3K and Akt,by upregulating the expression of CAV-1.更多还原
