OM-APS复方及其组分对新城疫疫苗免疫效果的影响
Effects of oxymatrine-astragalus polysaccharide compound and their components on immune response to newcastle disease vaccine in chicken
【摘要】 为观察OM-APS(氧化苦参碱-黄芪多糖)及其组分对新城疫疫苗免疫效果的影响,将14日龄非免疫健康罗曼蛋公鸡660羽随机均分为11组,第1~9组分别为免疫+OM-APS、OM和APS高、中、低剂量药物组,第10组为免疫对照组,第11组为不进行免疫和药物处理的空白对照组。除空白对照组外均用NDV-Ⅳ系疫苗滴鼻、点眼免疫,28日龄二免。在首次免疫的同时,1~9组分别口服药物,疫苗对照组和空白对照组口服等量生理盐水,每天1次,连续3 d。分别于14、21、28、35和42日龄采血,用MTT法测定T淋巴细胞增殖的变化,用β-微量法测定血清抗体效价变化。于14、28、42日龄采血分离血清,用ELISA方法检测IL-2和IFN-γ的浓度。结果表明:与疫苗对照组比较,高、中剂量的OM-APS、OM和APS均能显著提高血清抗体效价,促进T淋巴细胞增殖,增加血清IL-2和IFN-γ的含量,OM-APS在雏鸡多个日龄段效果优于OM和APS。以上结果表明,OM和APS经组方后可以提高新城疫疫苗的免疫效果。
【Abstract】 In order to figure out the effects of OM-APS(oxymatrine-astragalus polysaccharide) compound and their components on immune response of ND(newcastle disease) vaccine,six hundred and sixty 14-day-old non-immunized Roman cocks were randomly divided into 11 groups.Groups 1 to 9 were treated with OM-APS,OM and APS at high dose,medium dose,and low dose,respectively,and group 10 was treated with vaccine and group 11 was blank control.Groups 1 to 10 were inoculated with NDV-IV vaccine through intraocular-nasal route,repeated vaccination at day 28.At the same time of the first vaccination,the chickens in groups 1 to 9 were given the compounds through oral administration,and the chickens in group 1 and blank control group were given physiological saline,once a day for three days.On days 14,21,28,35 and 42,peripheral T lymphocyte proliferations and serum antibody titers were determined by MTT method and β-microscopic method respectively.On days 14,28 and 42,concentrations of IL-2 and IFN-γ of serum were determined by double antibody sandwich ELISA method.Tymphocyte proliferation,antibody titer and cytokines of chickens in the groups treated with high-and medium-dose of OM-APS,OM and APS were obviously higher than those in vaccine control group.At same ages of chickens,OM-APS performed better than OM and APS improving immune response to NDV-IV vaccine in chicken.It was indicated that the immunity of newcastle disease vaccine could be improved by OM-APS compound.
