鸡IGFBP-3基因C1087T位点遗传多态性及其对京海黄鸡体重和产蛋性能的遗传效应
Genetic polymorphism of chicken IGFBP-3 gene at C1087T and its association with body weight and productive performance of Jinghai yellow chicken
【摘要】 为了研究京海黄鸡、AA鸡、鹿苑鸡、边鸡4个鸡品种胰岛素样生长因子结合蛋白质-3基因(IGFBP-3)第2外显子多态性及其对京海黄鸡生长和繁殖性能的影响,采用PCR-RFLP技术和DNA测序技术对4个鸡种进行多态性检测,并将发现的单核苷酸多态性(SNP)与京海黄鸡部分经济性状进行关联分析。结果显示,MspⅠ酶切位点C1087T在4个鸡品种中基因型分布趋势一致,该位点属于中度多态信息含量位点,AA鸡多态信息含量最低(0.27)。χ2适合性检验结果表明除京海黄鸡外其余3个鸡品种C1087T位点均处于Hardy-Weinberg平衡状态(P>0.05)。关联分析结果显示,京海黄鸡CT基因型和TT基因型个体的300日龄体重显著大于CC基因型(P<0.05)。京海黄鸡TT基因型个体300日龄和66周龄产蛋数显著高于CC基因型个体(P<0.05),TT基因型个体的300日龄产蛋数和66周龄产蛋数分别为111.94和186.80个。由此初步推断MspⅠ位点对京海黄鸡生长和繁殖性能有一定的影响,TT型为增加产蛋数的优势基因型,该位点在生产上可以作为种鸡选育的候选分子标记进行选种。
【Abstract】 To reveal the polymorphism of the IGFBP-3 gene exon 2 among Jinghai yellow chicken,AA chicken,Luyuan chicken and Bian chicken,and its relationship with growth and reproductive performance in Jinghai yellow chicken,PCR-RFLP and DNA sequencing were used to detect the single nucleotide polymorphisms(SNPs) of IGFBP-3 exon 2,and the correlations of SNPs with economic important traits in Jinghai yellow chicken were analyzed.The result demonstrated that the MspⅠloci had the same gene frequency trend in the four chicken breeds,and belonged to the loci with the moderate polymorphism information content(PIC).The PIC of AA chicken was the lowest(0.27).The χ2 test revealed that locus C1087T in three chicken breeds except Jinghai yellow chicken were in Hardy-Weinberg equilibrium(P>0.05).Correlation analysis showed that the body weight of 300-day-old CT-type and TT-type Jinghai yellow chicken was significantly greater than those of CC-type(P<0.05),the egg production of TT-type Jinghai yellow chicken was significantly higher than that of CC-type at 300-day old and 66-week old(P<0.05),and at day 300 and week 66 the egg numbers of TT-type Jinghai yellow chicken were 111.94 and 186.80,respectively.Locus C1087T is likely to play a certain role in promoting the growth and reproductive performances,and TT genotype is the predominant genotype for egg-laying.The locus could be used as a candidate molecular marker for chicken breeding.
