北京市密云县野栖鼠感染田鼠巴贝西虫的检测

【摘要】 针对2012年6月于北京市密云县辖区捕获的15只野栖鼠,检测其巴贝西虫感染状况。通过制备薄血涂片及采集心、肺组织样本提取基因组,结合镜检及聚合酶链式反应确定感染阳性样本,对巴贝西虫核糖体小亚基编码基因虫种特异片段进行扩增并测序,通过序列对比及进化树构建对野栖鼠感染的巴贝西虫进行同源性分析及虫种鉴别。本次检测的野栖鼠中1份社鼠Niviventer confucianus血液镜检阳性,对应的心、肺组织样本检测到巴贝西虫虫种特异DNA片段,同源进化分析表明,该虫株与我国浙江、福建、台湾地区的田鼠巴贝西虫分离株具有高度同源性,属于田鼠巴贝西虫Babesia microti。北京市密云县野栖鼠首次被证实存在感染田鼠巴贝西虫的情况。

【Abstract】 In June 2012,15 rodents captured in Miyun district of Beijing were detected for the infection of Babesia parasites. Thin blood smears were prepared with Giemsa’s stain for determining the intraerythrocytic Babesia parasites and the samples of heart and lung tissues in suspicious rodents were detected for the species specific segments of 18S rRNA coding genes of Babesia spp. by using polymerase chain reaction( PCR) method. One N. confucianus was found infected with Babesia microti by directly observation on thin blood smear and PCR test and subsequent sequences. High homology of the strains with those Zhejiang,Fujian and Taiwan strains in BLAST and phylogenetic analysis revealed that the species of Babesia parasite might be B. microti. For the first time,Babesia microti were detected among rodents in Miyun district of Beijing,which indicates the potential risk of human health.
