人胚胎生殖细胞移植治疗猪心肌梗死

【摘要】 目的:研究人胚胎生殖细胞(EG细胞)移植猪心肌梗死区存活及心肌再生作用。方法:取5~10周人胚胎生殖腺嵴,采用组织块培养法体外培养获得人胚胎生殖细胞。取太湖小型猪2只,分移植组和对照组,开胸结扎冠状动脉左前降支制作心肌梗死模型。术后2周处死猪,取出心肌组织,以鼠抗人细胞核抗体MAB1281作为示踪剂,免疫组织化学检测移植细胞的存活情况;并观察转录因子GATA-4、连接蛋白Cx43、心肌转录因子NKx-2.5和心肌肌钙蛋白T(cTnT)在移植猪心组织的表达。结果:成功地建立猪急性心肌梗死模型,结扎冠脉后可见心电图典型的心肌梗死变化;免疫组织化学显示移植组MAB1281阳性细胞,GATA-4、Cx43、Nkx2.5和cTnT均阳性表达。结论:人EG细胞移植于此模型的心肌组织中不仅存活,而且能向心肌细胞分化,具有心肌再生能力。

【Abstract】 Objective:To study the effect of human embryonic germ cells(hEGCs) on myocardial regeneration.Methods:The gonadal ridges from human embryos aged 5-10 weeks were taken to serve as transplanted cells.Two swine were equally divided into a transplanted group and a control group.A model of myocardial infarct in the swine was created by the ligation of the anterior descending branch of the left coronary artery.The hEGCs were transplanted through epicardium,these pigs were sacrificed 2 weeks after transplantation.Immunohistochemistry was used to detect the expression of anti-human cellular nuclei MAB1281 in order to localize the implanted cells.Similarly,the expression of transcription factor GATA-4,Cx43,Nkx2.5and cTnT were detected in the transplanted cells.Results:The acute myocardial infarction model of pigs was successfully established.The typical electrocardiogram changes of myocardial infarction were showed after coronary artery ligation.In the transplanted group,transcription factor GATA-4,connexin Cx43,transcription factor Nkx 2.5 and cardiac specific protein cTnT were showed a positive expression in the anti-human nuclei antibody MABl281-positive cells.The control group were negative.Conclusion:Human EGCs cells transplanted into pigs’ myocardium through epicardium can live and differentiate into cardiomyocytes,and integrate with the host myocardial cells.
