同系异体移植当归多糖动员的外周造血干细胞治疗小鼠弥漫性酒精性肝损伤
Treatment of syngeneic and allogeneic transplantation of Angelica polysaccharide mobilized peripheral blood hematopoietic stem cells on mice with diffuse alcoholic liver injury
【摘要】 目的:探讨当归多糖动员的外周造血干细胞对小鼠酒精性肝损伤的作用。方法:采用磁珠分选技术,提取当归多糖动员的正常小鼠外周血造血干细胞,将其注入小鼠酒精性损伤的肝,观察经当归多糖动员的造血干细胞对小鼠酒精性肝损伤的作用。结果:注射当归多糖动员出的造血干细胞后,小鼠酒精损伤的肝组织在48 h左右,肝细胞得到明显修复,肝小叶的正常结构增多,小鼠生存期延长。结论;当归多糖可促进骨髓造血干细胞增殖并向外周动员;当归多糖动员至外周的造血干细胞对小鼠酒精性肝损伤有显著治疗作用;同系异体之间干细胞可进行移植。
【Abstract】 Objective:To explore the effect of angelica polysaccharide(APS) mobilized peripheral hematopoietic stem cells on alcoholic hepatic injury in mice.Methods:Using magnetic bead sorting technique,the peripheral blood hematopoietic stem cells were extracted from APS mobilization in normal mice to inject into the mouse with alcoholic injury of the liver.The effect on alcoholic hepatic injury in mice was observed.Results:After injecting hematopoietic stem cells of APS mobilization to alcohol mice,the liver cells were significantly repaired in 48 hours;the hepatic lobule of the normal structure increased;and the survival time of mice was prolonged.Conclusion:APS can promote the proliferation of hematopoietic stem cells in bone marrow and peripheral mobilization;APS mobilization to the peripheral hematopoietic stem cells on alcoholic hepatic injury in mice has a significant therapeutic effect;and stem cells can be transplanted between syngeneic and allogeneic.
