肿瘤细胞分泌的VEGF-C和CCR7在恶黑淋巴道转移中的作用

【摘要】 目的研究血管内皮生长因子C(VEGF-C)和C-C家族趋化因子受体7(CCR7)与恶性黑色素瘤(恶黑)淋巴管浸润、淋巴结转移的关系及其预后价值。方法免疫组化方法检测56例恶黑组织中VEGF-C和CCR7的表达,淋巴管内皮细胞透明质酸受体1(LYVE-1)标记肿瘤的淋巴管,Kaplan-Meier法进行生存检验,应用Cox比例危险度模型筛选与恶黑预后有关的指标。结果肿瘤细胞胞浆中可检测到VEGF-C和CCR7的表达。CCR7表达与VEGF-C表达和淋巴管浸润有关,CCR7和VEGF-C协同增加淋巴管浸润,CCR7表达与淋巴结转移及预后无关。结论在恶黑组织中VEGF-C和CCR7诱导肿瘤细胞侵入到淋巴管,但CCR7和淋巴管浸润不能作为恶黑的预后指标。

【Abstract】 Objective To study the relationship of vascular endothelial growth factor-c(VEGF-C) and C-C chemokine receptor 7(CCR7) expressions with lymphatic invasion, lymph node metastasis and the prognosis in malignant melanoma. Methods Expressions of VEGF-C and CCR7 were immunohistochemically evaluated in 56 malignant melanoma patients.LYVE-1 was selected as a lymphatic endothelial cell marker. Survival was analyzed by the Kaplan–Meier method. Indicators related to the prognosis of malignant melanoma were screened using Cox’s proportional hazards models. Results The expressions of both VEGF-C and CCR7 are detected in the cytoplasm of the malignant cells. CCR7 expression was positively correlated to the expression of VEGF-C and lymphatic invasion, CCR7 and VEGF-C synergistically enhanced lymphatic invasion, but no correlation between CCR7 expression and lymph node metastasis. Conclusion CCR7 and VEGF-C might be involved in tumor lymphatic invasion,simultaneously, but CCR7 and lymphatic invasion could not be markers for evaluation of clinical prognosis in patients with malignant melanoma.
