目的:建立一种检测微量蛋白质的滚环扩增技术(rolling circle amplification,RCA)。方法:以血管内皮生长因子为研究对象,利用链亲和素做桥接,将免疫反应和RCA技术进行有效的结合,构建一种新型的蛋白质检测方法。结果:通过优化实验条件,对血管内皮生长因子检测的灵敏度达到10fM,大大超过传统的ELISA方法。结论:RCA技术联合免疫反应能够高特异性、高灵敏度地检测低丰度蛋白质,具有广阔的临床应用前景。
