响应面法优化畜禽用凝结芽孢杆菌发酵培养基
Study on the Fermentation Medium of Bacillus coagulans Applied to Animal by Response Surface Method
【摘要】 为了获得最佳发酵培养基,提高凝结芽孢杆菌液体发酵水平,在单因素试验优化基础上,采用Plackett-Burman设计确定淀粉、大豆粉和鱼粉混合物(质量比2∶1)和磷酸氢二钾为凝结芽孢杆菌液体发酵水平的显著影响因素,通过最陡爬坡路径试验、中心组合设计和响应面分析法确定最优培养基组成。试验结果表明,最优培养基组成为淀粉10.6g/L、大豆粉13.5g/L、鱼粉6.7g/L、磷酸氢二钾2.72g/L、磷酸二氢钠0.312g/L、碳酸钙0.2g/L、一水硫酸锰0.169g/L、硝酸钠0.34g/L和七水硫酸镁0.74g/L。在最佳培养基组成条件下,液体发酵活菌数达67×108 CFU/mL。
【Abstract】 Researchs on fermentation medium of Bacillus coagulans strain applied to animal on singlefactor experiment basis have been done.Three important factors of starch,mix of soybean meal and fish meal(mass ratio was 2to 1)and dipotassium hydrogen phosphate,which can influence the living bacteria amount,were screened through Plackett-Burman design and test.The results showed that the optimal medium was starch 10.6g/L,mix of bean flour 13.5g/L and fish meal 6.7g/Land dipotassium hydrogen phosphate 2.72g/L by using the steepest ascent path,center combination of design and response surface analysis.Under the optimized medium,the living bacteria amount of fermentation broth can reach 67×108 CFU/mL.
