Box-behnken效应面法优化补骨脂素中空黏附微球的制备

【摘要】 目的筛选制备最佳补骨脂素中空黏附微球的工艺处方。方法采用Box-behnken实验设计优化、筛选制备最佳补骨脂素中空黏附微球的工艺处方,并应用Design-Expert(8.0.4,试用版)软件分析试验结果。结果所得优化后的处方是投药量为12.36%(Y1)、乙基纤维素/EudragitEPO(EC/Eu)比值为2.27、PVA的浓度为1.53%,制备的补骨脂素中空黏附微球的药物含量为9.18%(Y1),6 h的百分释放率为76.15%(Y2),球形度为80.97%(Y3)。结论所用方法筛选制备补骨脂素中空黏附微球的最佳处方是可行的。

【Abstract】 OBJECTIVE To optimize the formulation parameters of psoralen loaded hollow-bioadhesive microspheres.METHODS Box-behnken experimental design was employed to statistically optimize the formulation parameters of psoralen loaded hollow-bioadhesive microspheres.A three-factor,three-level design of experiment with response surface methodology was run to evaluate the main and interaction effect of several independent formulation variables that were included.RESULTS The optimized microspheres were obtained with a drug loading(Y1) of 9.18%,cumulative drug release in 6 h(Y2) of 76.15%,and spherisity(Y3) of80.97%,respectively.When the drug fed amount(X1),Ec∶Eu(X2),and PVA(X3) values were 12.36%,2.27 and 1.53%,respectively.CONCLUSION The results indicated Box-behnken experimental design facilitates the formulation and optimization of psoralen loaded hollow-bioadhesive microspheres.
