视神经炎小鼠视网膜形态结构和视觉功能的变化
Morphological Changes of Retinal and Visual Function of Mice with Experimental Optic Neuritis
【摘要】 目的研究视神经炎小鼠发病过程中视网膜形态结构和视觉功能的变化。方法 50只C57BL/6小鼠随机分为对照组(10只)和实验性自身免疫性脑脊髓膜炎(experimental autoimmune encephalomyelitis,EAE)组(40只),EAE组应用髓鞘少突胶质细胞糖蛋白(myelin oligodendroglia glycoprotein,MOG)35-55多肽加完全弗氏佐剂皮下注射免疫建立视神经炎模型。对照组和EAE组小鼠分别于免疫当天,免疫后第7、14、20、30天,进行闪光视觉诱发电位(flashvisual evoked potential,F-VEP)和闪光视网膜电图(flash electroretinogram,F-ERG)检查,采用勒克斯坚牢蓝(Luxol fast blue,LFB)染色评估视神经的髓鞘脱失程度;采用原位末端转移酶标记技术(terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling,TUNEL)检测视网膜神经节细胞凋亡并计算其凋亡指数。结果在免疫后第7天,F-ERG中b波振幅有降低趋势,潜伏期延迟,和对照组比较有统计学差异(P<0.05);F-VEP结果和对照组比较无统计学差异(P>0.05);在免疫后第14天,视神经开始出现不同程度的髓鞘脱失;F-VEP中P100波潜伏期明显延迟,振幅明显降低,和对照组比较有统计学差异(P<0.05);TUNEL检测,EAE组有明显的视网膜神经节细胞(ratinal ganglioncells,RGCs)凋亡,与对照组比较有统计学差异(P<0.05)。凋亡指数随病程的延长而升高。结论F-ERG和F-VEP可以在一定程度上反映视神经炎临床前期的视觉功能,为评估病情程度提供依据;早期RGCs的凋亡提示,对视神经炎进行及时神经保护治疗是必要的。
【Abstract】 Objective To explore the changes of morphometry of retina and visual function of mice with experimental optic neuritis.Methods A total of 50 C57BL/6 mice were divided into control group(10 mice) and experimental autoimmune encephalomyelitis(EAE) group(40 mice).Myelin oligodendrocyte glycoprotein(MOG)35-55peptide in CFA were injected subcutaneously in C57BL/6 mice to induce optic neuritis and EAE.Flash visual evoked potentials(F-VEP) and flash electroretinogram(F-ERG) tests were performed in two groups.Luxol fast blue(LFB) stain of consecutive sections of optic nerve was utilized to assess inflammation,demyelination and axonal pathology on 0,7,14,20,30 days after immunity.By terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling(TUNEL) technique the apoptosis of retinal ganglion cells(RGCs) were measured and the apoptosis index were calculated.Results At 7 days post immunization,the amplitude of F-ERG b wave was decreased,the latency was delayed in EAE group,which was significantly different from that of control group(P<0.05).There was no difference of F-VEP in two groups(P>0.05).At 14 days after immunization,the latency of F-VEP P100 wave was significantly delayed,and the magnitude was decreased in EAE group,which was significantly different from that of control group(P<0.05).Obvious apoptosis of RGCs was observed in EAE group,the apoptotic index of RGCs was significantly higher than that in control group(P<0.05) and increased with the prolonger of the disease course.Conclusion F-VEP and F-ERG can reflect the function of vision in optic neuritis in preclinical phase and can supply reference to assess the state of illness.The apoptosis of RGCs in early period indicated that the timely optic nerve protection treatment is necessary.
