燃煤型氟中毒大鼠血清骨碱性磷酸酶及Ⅰ型胶原羧基端前肽的变化

【摘要】 目的观察骨碱性磷酸酶(BALP)及Ⅰ型胶原羧基端前肽(CⅠCP)在燃煤型氟中毒大鼠中的表达变化,探讨骨形成在氟中毒骨病变中的地位。方法将断乳4周的SD大鼠160只随机分为对照组、低氟组、中氟组、高氟组(饲料氟:6.30、9.56、15.89、23.00 mg/kg),食用病区煤烘玉米,复制燃煤型氟中毒动物模型。分别于染氟后30、90、180 d以ELISA法检测血清BALP和CⅠCP水平。结果染氟30和90 d时各组动物血清BALP水平差异无统计学意义,180 d时中氟组、高氟组BALP均高于对照组和低氟组(P<0.01)。染氟30 d时高氟组血清CⅠCP水平低于对照组(P<0.05);90 d时中氟组和高氟组血清CⅠCP水平均高于对照组(P<0.05),且有随着剂量增加而增加的趋势;180 d时各组动物血清CⅠCP水平高于对照组(P<0.01),且有随着剂量增加而增加的趋势。结论氟可以使大鼠骨形成加速,且氟对CⅠCP表达的影响较BALP显著。

【Abstract】 Objective To observe the changes of BALP and CⅠCP in rats with coal-burning fluorosis and to explore the role of osteogenesis in skeletal fluorosis.Methods A total of 160 SD rats weaned for four weeks were randomly divided into control group,low,moderate,high-dose group,respectively.All of the rats were fed with the corn(fluoride level: 6.30,9.56,15.89,23.00 mg/kg) roasted with the coal from the endemic fluorosis area.After treatment for 30 days,90 days and 180 days,levels of BALP and C Ⅰ CP in serum were measured with ELISA.Results After treatment for 30 days and 90 days,no significant deference was seen in BALP among the four groups.For 180 days,the BALP levels in high-dose and medium-dose group were higher than that of control group(P<0.01).After treatment for 30 days,value of CⅠCP in high-dose group was lower than that of control,For 90 days,the levels of CⅠCP in high-dose group and moderate-dose group were higher than that of control,and for 180 days,the levels of C Ⅰ CP in all exposure groups were higher compared with the control.Conclusion Fluoride can enhance the osteogenesis in rats,and the effect of fluoride on CⅠCP is more obvious than on BALP.
