清开灵注射液对小鼠TB细胞增殖与功能的影响

【摘要】 目的:研究清开灵注射液在体内对正常小鼠胸腺及脾脏指数,迟发型过敏反应(DTH)及T、B细胞活化增殖的影响,探讨其免疫调节作用及机制。方法:将小鼠随机分为正常对照组、DTH对照组、清开灵注射液低、中、高三个剂量组及阳性药物组。各组小鼠给予不同浓度的清开灵注射液,阳性药物或生理盐水腹腔注射后,对各组小鼠胸腺及脾脏指数,DTH反应强度以及T、B细胞增殖活化能力进行比较。结果:清开灵注射液大、中两个剂量组的小鼠胸腺指数明显低于正常对照组,存在极显著性差异(P<0.01),而清开灵注射液对小鼠脾脏指数无明显影响(P>0.05);三个剂量的清开灵注射液均能明显抑制DTH反应,与DTH组相比,差异显著(P<0.05);清开灵注射液三个剂量组均可明显降低正常小鼠由ConA刺激的T细胞非特异性增殖反应,其中以大剂量组作用最为明显,和正常组相比,差异极显著(P<0.001)。而对由LPS刺激的B细胞非特异性增殖反应,则无显著性影响(P>0.05)。结论:清开灵注射液体内可有效抑制T细胞的活化增殖及功能从而调节机体的免疫反应。

【Abstract】 Objective: To study the influence of Qingkailing injection on the mouse spleen index and thymus index,delayed type hypersensitivity(DTH) in vivo,and the proliferation of mouse spleen T and B lymphocytes,and to explore the regulatory effects and mechanisms of Qingkailing injection on immune system.Method: Mice were divided into six groups: normal group,DTH model group established by stimulation with DNFB,three different concentration Qingkailing injection groups and the positive control drug group.After different doses of Qingkailing injection were used for 10 days,the spleen index and thymus index was examined Finally concanavalin A was added to mouse spleen lymphocytes to activate T lymphocytes and LPS to activate B lymphocytes.MTT method was used to observe the effects of different groups on mouse spleen T and B lymphoctes proliferation.Result: The thymus indexes of high and media concentration Qingkailing injection groups were obviously lower than normal group(P <0.01),however the spleen indexes of Qingkailing injection groups were not obviously different with normal group(P >0.05).All the three different concentration of Qingkailing injection could obviously inhibit the delayed type hypersensitivity(P >0.05).Qingkailing injection of different concentration could obviously inhibit the proliferation of spleen T lymphocytes activated by concanavalin A(P <0.001),but it could not influence the proliferation of spleen B lymphocytes activated by LPS(P >0.05).Conclusion: Qingkailing can regulate immune reaction by obviously inhibiting the activation and proliferation of T lymphocytes in vivo.
