祁木香提取物的药效学研究
Pharmacodynamic study on extract from radix inulae
【摘要】 目的研究祁木香提取物的药效学作用。方法①采用碳末法,将昆明种小鼠随机分为空白对照组、阳性对照组、祁木香低剂量组、祁木香中剂量组和祁木香高剂量组,各组10只,禁食不禁水后,空白对照组给予纯净水,阳性对照组给予吗丁啉(5×10-3g/kg),各实验组灌胃给予祁木香提取物,每日1次,连续5 d。末次给药30 min后,灌服碳末混悬液0.2 mL/只,20 min后颈椎脱臼法处死小鼠,从幽门至碳末前沿为"碳末推进距离",计算碳末推进百分率,观察祁木香提取物对小鼠小肠推进的影响。②采用小鼠热板法,将小鼠随机分为5组,每组10只,分别为空白对照组、阳性对照组、祁木香低剂量组、祁木香中剂量组、祁木香高剂量组。1 d后开始给药,在给药之前每只小鼠再测一次痛阈值,将此痛阈值与该只小鼠筛选时的痛阈值取平均值,以此平均值作为该只小鼠在给药前的正常痛阈值。空白对照组给予纯净水,阳性对照组给予阿司匹林,各实验组灌胃给予祁木香提取物,每日1次,连续给药5 d。分别在末次给药后30、60、90 min时间段用热板仪测定小鼠出现舔足的时间,超过60 s不舔足者按60 s计。比较用药前后各组出现舔足时间作为动物疼痛反应的指标。③冰醋酸致小鼠扭体反应法,将小鼠随机分为5组,每组10只,分别为空白对照组、阳性对照组、祁木香低剂量组、祁木香中剂量组、祁木香高剂量组。空白对照组给予纯净水,阳性对照组给予阿司匹林,各实验组灌胃给予祁木香提取物,每日1次,连续给药5 d。末次给药30 min后,每只小鼠均腹腔注射0.8%冰醋酸0.2 mL,随即观察出现扭体反应的时间(扭体潜伏时间)和20 min内出现扭体反应的次数。计算药物对扭体反应的抑制率,观察祁木香提取物对疼痛的抑制作用。结果碳末法实验中,祁木香提取物祁木香中、高剂量组小肠的推进率高于空白对照组(P<0.01);在热板法实验中,祁木香提取物与空白对照组比较,祁木香低剂量组与空白对照组比较差异无统计学意义(P>0.05);祁木香中剂量组在给药后60 min能显著提高小鼠对热反应的痛阈值(P<0.05),祁木香高剂量组在给药后30、60、120 min时间段对小鼠热反应的痛阈值均有提高(P<0.01);在扭体实验中,祁木香提取物祁木香高剂量组能显著抑制冰醋酸诱发小鼠扭体反应(P<0.05),抑制率为42%。结论祁木香提取物具有较好的促进小肠推进作用和镇痛作用。
【Abstract】 Objective To study the pharmacological effects of exact from radix inulae. Methods The effects of small intestinal peristalsis of mice were observed by using carbon powder method. The Kunming mice were divided into control group,positive control group,low dose group,middle dose group and high dose group. Control group received purified water,positive control group received Domperidone(5 × 10- 3g / kg),each treatment group received different dose exact from radix inulae,once a day for five days. After 30 min the last administration,carbon powder method was proceeded. The percentage of carbon propelling was calculated for evaluating effects of extract from radix inulae on small intestinal peristalsis. While the analgesic effect of exact from radix inulae was observed by using pain models induced by heatplate and acetic acid-induced writhing at the same time. 50 Kunming mice were divided into control group,positive control group,low dose group,middle dose group and high dose group. Control group received purified water,positive control group received Aspirin,each treatment group received different dose exact from radix inulae,once a day for five days. After 30 min the last administration,heatplate method was proceeded.And after 30 min the last administration,acetic acid- induced writhing method was proceeded. Initial time of mice licking,writhing latency and writhing number were determinated. Results The propulsion rate of small intestinal in middle dose group and high- dose group was significantly higher than that in control group in carbon powder experimental(P < 0. 01). High- dose group significantly reduced the aching reaction induced by heat and acetic acid inhibition rate was 42%; and middle dose significantly reduced the aching reaction induced by heat. Conclusion s The extract from radix inulae has positive modulation on mouse’s intestinal movements and significant analgesic effect.
