无细胞百日咳脱毒PT抗原超滤工艺研究
Study of Ultrafiltration Process on Detoxified PT Antigen of Acellular Pertussis
【摘要】 通过超滤去除无细胞百日咳脱毒PT抗原的色素、脱毒剂等小分子,考察了超滤装置、超滤液类型及其pH值、超滤膜孔径对超滤后制品外观、蛋白回收率的影响。结果表明,使用30kD孔径的膜式超滤器,用pH值为7.8的含0.5mol·L-1 NaCl的PB缓冲溶液对无细胞百日咳脱毒PT抗原进行超滤,能有效去除色素、脱毒剂等小分子,超滤后制品物理外观良好,蛋白回收率接近80%。
【Abstract】 Using ultrafiltration to remove pigment,detoxification agent of detoxified PT(Pertussis toxin) antigen of acellular pertussis was studied.The effects of ultrafiltration apparatus,type and pH value of ultrafiltration solution,and pore size of ultrafiltration membrane on product appearance and the protein recovery rate were investigated.The results showed that pigment and detoxification agent of detoxified PT antigen could be removed efficiently by using membrane ultrafilter of 30 kD pore size,PB buffer solution containing 0.5 mol·L-1 NaCl with pH value of 7.8.After ultrafiltration,the product appearance was well,and the protein recovery rate reached nearly 80%.
