高效耐高温石油降解菌降解石油的促进因子研究
Study on the Promotion Factors for Degradation of Petroleum by High Efficient Thermostable Petroleum Degrading Bacteria
【摘要】 将耐高温石油降解菌UM6-3菌悬液接种于100mL石油MSM中,在47.5℃下振荡培养6d。分别考察环境因子中的初始pH值、石油含量、振荡器转速,营养平衡因子中的氮源、氮磷比,生物因子中的接种量、酵母粉添加量对石油降解的促进作用。基于单因素实验结果,设计初始pH值、接种量、氮磷比、石油含量、振荡器转速的5因素4水平正交实验,结果表明,影响石油降解的主要因子是振荡器转速和接种量,初始pH值的影响最小;UM6-3降解石油的最佳促进因子为:初始pH值7,接种量4mL,氮磷比5∶1,石油含量1%,振荡器转速140r.min-1。
【Abstract】 UM6-3 Bacteria were seeded in the 100 mL of petroleum MSM,and cultured for 6 d with shaking under the temperature of 47.5 ℃.The environmental factors(such as initial pH value,petroleum content,rotation speed),nutritional balance factors(such as nitrogen source,nitrogen-phosphorus ratio) and biological factors(inoculum amount and yeast powder addition amount),which may promote the effect of petroleum degradation,were studied.Based on the single factor experiment results,the L16(54) orthogonal experiment was designed for the factors including initial pH value,inoculum amount,petroleum content,nitrogen-phosphorus ratio and rotation speed.The results showed the main factors affecting the petroleum degradation were rotation speed and inoculum amount,initial pH value was inferior.For UM6-3,the optimum promoting factors were as follows:initial pH value of 7,inoculum amount of 4 mL,N∶P of 5∶1,petroleum content of 1%,rotation speed of 140 r·min-1.
