辐照灭菌工艺对熟地黄中梓醇含量的影响
Studies on the effect of radiation sterilization technology to the content of catalpol in Prepared Rehmannia glutinosa
【摘要】 目的考察60Co-γ辐照灭菌工艺对熟地黄中梓醇含量的影响。方法采用HPLC法对熟地黄中的梓醇进行定性鉴别及含量测定,流动相:乙腈-甲醇-0.1%磷酸溶液(10∶90∶1);柱温:30℃;检测波长:215nm;流速:1.0mL·min-1。结果梓醇在5~100μg·mL-1范围内呈良好线性关系(r=0.9998),辐照前后样品中梓醇的含量变化在95.07%~98.86%之间。结论 60Co-γ辐照灭菌工艺对熟地黄中梓醇含量无明显影响,可用于熟地黄的灭菌。
【Abstract】 OBJECTIVE To study the content variation of catalpol in prepared Rehmannia glutinosa radiated by60Co-γ or not. METHODS The catalpol was identified and the content of catalpol was detected by HPLC in215nm: the mobile phase was a mixed liquid with acetonitrile-methanol-water( 10∶ 90∶ 1),the column temperature was30℃ and the flow rate was 1. 0mL · min-1. RESULTS The content of catalpol in all samples was between95. 07% ~ 98. 86% and the method was linear within 5 ~ 100μg·mL-1which means it can be used to test the content of catalpol in prepared Rehmannia glutinosa,CONCLUSION The content of catalpol in prepared Rehmannia glutinosa radiated by60Co-γ is not significant decline,so it is suitable to sterilization for prepared Rehmannia glutinosa by the60Co-γ radiation sterilization technology.
