HL60/VCR耐药细胞荷瘤鼠模型的建立
Establishment of the implanted animal model of human multidrug resistant cell line HL60/VCR in BALB/c-nu/nu mice
【摘要】 目的建立白血病HL60/VCR耐药细胞株移植动物模型方法。方法将生长状态良好的耐药细胞株HL60/VCR1.0×107/0.2ml接种到BALB/c-nu/nu裸小鼠皮下,定期观察裸鼠生长状况,测量移植瘤大小,留取标本检测不同脏器肿瘤负荷及多药耐药基因MDR1表达情况。结果 2.0×106HL60/VCR细胞皮下接种BALB/c-nu/nu裸小鼠,成瘤率为67.7%,免疫组化检测荷瘤裸鼠肝脏和脾脏均可见巢状分布白血病细胞,PCR检测成瘤组织表达MDR1基因。结论皮下移植可成功建立耐药细胞株荷瘤鼠模型,为研究多药耐药细胞株动物体内生物学行为及逆转多药耐药提供了动物模型。
【Abstract】 Objective To establish the implanted animal model of human multidrug resistant cell line HL60/VCR in mice. Methods The concentration of Ishikawa cells in good condition were adjusted to 1. 0 × 107/0. 2ml were subcutaneously injected into BALB / c-nu / nu mice. Growing condition of tumor was observed,the specimens were taken for pathology examination and tumor marker in the body was monitored by PCR. Results A subcutaneous implanted tumor model of HL60 / VCR was established successfully in BALB / c-nu / nu mice. The formation ratio was 67. 7%. Immunohistochemistry showed tumor cells in liver and spleen,the expression of MDR1 gene was detected by PCR. Conclusion Subcutaneous transplantation can be used to establish an animal model of HL60 / VCR tumor cells in BALB / c-nu / nu mice successfully. This may be useful for study of behavior of multidrug resistance cell line in vivo and reversal of multidrug resistance.
