同源异体间充质干细胞上清液干预大鼠佐剂性关节炎

【摘要】 目的:探讨同源异体骨髓来源的间充质干细胞上清液对大鼠佐剂性关节炎的预防作用。方法:培养扩增SD大鼠骨髓间充质干细胞,收集培养间充质干细胞48 h的上清液;将健康雄性SD大鼠右后足跖皮内一次性注射弗氏完全佐剂0.1 mL/只制作佐剂性关节炎模型,造模后行间充质干细胞上清液干预;造模前及造模后第7、14、21和28天测量大鼠体重和后足爪体积,同时观察大鼠生存状况;并于干预后第28天摄四肢关节X片并做病理检查。结果:间充质干细胞上清液干预组大鼠的原发性炎症和继发病变均不明显,关节X片检查和病理检查均未见明显异常;干预组与模型组大鼠相比,体重、足爪体积差异有统计学意义(P<0.05)。结论:间充质干细胞上清液可有效干预弗氏完全佐剂诱导的大鼠佐剂性关节炎的进展。

【Abstract】 Objective: To explore the effect of culture supernatant of marrow mesenchymal stem cells(MSCs) in prevention of adjuvant arthritis(AA) in rats and its mechanism.Methods: Supernatant of MSCs cultured for 48 hours was collected.Rat disease model was made by injection intracutaneously with 0.1 ml of Freund’s complete adjuvant into right rear paw of rats.The model rats in intervention group were given 2 mL of MSCs supernatant intravenously in every 2 days,and 4 times in total.Model rats in control group were given L-DMEM culture medium instead.The rats’ weights and hind paw volumes were measured on the 1st,7th,14th,21stand 28thdays of the intervention respectively.And the rats’ living conditions were observed.On the 28th day of the intervention,X-ray examination and pathological examination were carried out on rats’ limb joints.Results: The primary inflammation and secondary lesions in rats of intervention group were not obvious,and joint X-ray and pathological examination showed no obvious abnormalities.Compared with model group,the differences of rats’ weight and paw volume were statistically significant(P <0.05).Conclusion: MSCs supernatant can intervene the progress of AA.
