【摘要】 目的体外探讨PADI4基因与白血病细胞分化的关系。方法检测HL60、K562、U937等9株不同白血病细胞株中PADI4在mRNA水平的表达情况,建立ATRA诱导HL60细胞分化模型,然后分别利用RT-PCR及WB技术检测PADI4基因在转录和翻译水平的表达变化。结果不同类型的白血病细胞株中PADI4表达情况不同。ATRA诱导HL60细胞后,在药物作用的96h内,随着药物作用时间的增加,在转录和翻译水平PADI4表达水平逐渐下调,与对照组相比差异具有统计学意义(P<0.01)。结论 PADI4与白血病细胞的分化有关,ATRA诱导HL60细胞分化后PADI4表达下调。更多还原

【Abstract】 Objective To explore the relationship of PADI4 gene with the differentiation of leukemia cells in vitro. Methods Detected the PADI4 expression at the mRNA level in different leukemia cells such as HL60、K562、U937 ect.Established the ATRA-induced HL60 cell model,detected the PADI4 expression at mRNA and protein levels by RT-PCR and WB method. Results Different leukemia cells kept different expression levels of PADI4 gene.PADI4 expression both at mRNA and protein levels gradually reduced along with the increased in the time of ATRA induced in 96h,and compared with the control group,the difference was statistically significant(P < 0.01). Conclusion The PADI4 gene affects the differentiation of leukemia cells,the expression levels of PADI4 gene will reduce in ATRA-induced HL60 cells.更多还原
