HP1188-IgY体内抗幽门螺杆菌感染的实验研究

【摘要】 目的研究抗幽门螺杆菌(H.pylori)HP1188蛋黄抗体(HP1188-IgY)在小鼠体内抗感染作用。方法用Western blot法分析HP1188-IgY抗原特异性。建立H.pylori感染Balb/c小鼠的动物模型,预防组在小鼠灌喂H.pylori菌液前灌喂不同剂量(1、3、5mg/mL)的HP1188-IgY,同时设PBS对照组、阴性对照组、阳性对照组。通过小鼠胃组织H.pylori培养、快速尿素酶实验和组织学检查观察H.pylori定植情况。结果 Western blot结果显示在相对分子质量30 000附近出现反应条带。阳性对照组8周后H.pylori的感染率为100%。HP1188-IgY剂量为3mg/mL和5mg/mL时,小鼠胃黏膜的H.pylori定植量减少,炎性反应减轻(P<0.05)。随HP1188-IgY剂量的增加,感染率降低。结论该实验成功制备了HP1188-IgY,在小鼠体内能阻止H.pylori的定植,为进一步制备预防H.pylori感染的口服制剂奠定了基础。

【Abstract】 Objective To study the effect of HP1188-IgY against Helicobacter pylori(H.pylori)in Balb/c mice.Methods The purity of HP1188-IgY was analyzed by Western blot.Before infection with H.pylori,Balb/c mice were administrated with diferent dosages(1,3,5mg/mL)of HP1188-IgY by using an oral feeding needle in the prevention group.The planting of bacteria in stomach was assayed by observing bacterial culture,rapid urease test and pathological section.Results Western blot exhibited the protein bands with molecular weight of 30 000.The total rate of infection in the positive control group was 100%after 8weeks.The infection rate of the prevention group(3and 5mg/mL)was much lower than that of the PBS control group at the same period of infection(P<0.05),and was negatively related to the addition of HP1188-IgY.Conclusion The HP1188-IgY was successfully constructed,which is able to inhibit the adhesive attraction of H.pylori in vivo and be used to prevent H.pylori infection.
